ScerGAL4VT001211 drives expression in VGlut1 positive mushroom body medial lobe arborizing neuron 4 and mushroom body medial lobe arborizing neuron 6. EcollexAGMR48B04 positive dopaminergic PAM neurons are in close proximity with ScerGAL4VT001211 expressing mushroom body medial lobe arborizing neurons 4 and 6 as shown by a positive GFP reconstitution across synaptic partners (GRASP) signal.

Inhibition (using shi1.UAS at 30-33[o]C) of Scer\GAL4VT001211 neurons in adults, only during aversive (CS+) odor re-exposure following training, prevents the aversive olfactory memory extinction that results from re-exposure; inhibition during non-aversive (CS-) odor re-exposure abolishes avoidance of the aversive (CS+) odor, compared to controls; there is no effect on memory performance when inhibition occurs between training and testing with no odor re-exposure.

Blocking synaptic transmission (using shi1.Scer\UAS at 32[o]C) from Scer\GAL4GMR21D02, Scer\GAL4VT001211, or Scer\GAL4GMR66C08 neurons significantly impairs memory performance compared to controls. With sucrose-reinforced appetitive conditioning or electric shock-reinforced aversive conditioning, immediate memory at 3 minutes post-conditioning is impaired (both training and testing at a restrictive 32[o]C temperature). Appetitive conditioning memory at 24 hours is also impaired (training at a permissive temperature of 23[o]C, testing at the restrictive temperature).

Blocking synaptic transmission (using shi1.Scer\UAS at 32[o]C) from Scer\GAL4VT001211 neurons significantly impairs memory performance (2 hours after appetitive or aversive conditioning) compared to controls; training is at a permissive temperature of 23[o]C, testing is at the restrictive 32[o]C temperature.

Blocking synaptic transmission (using shi1.Scer\UAS at 32[o]C) from Scer\GAL4GMR21D02 or Scer\GAL4VT001211 neurons significantly abolishes or reverses (depending on the odorant and concentration used) naive odor avoidance behavior, compared to controls.
