Description

The pHCl-2 gene has been deleted and replaced with the following sequence: a single attP site and a loxP cassette which itself contains an mFRT71 cassette followed by Scer\GAL4 coding sequence and a polyadenylation signal (this GAL4 sequence is non-functional). The mFRT71 cassette contains 1: the mCherry coding region tagged with a membrane-targeting signal (Tag:Myr(Unk)) and followed by a polyadenylation signal (this is expected to be expressed under the control of the endogenous pHCl-2 regulatory sequences), 2: a w+* marker.

pHCl-2pTV-Cherry homozygotes exhibit a developmental delay (following re-colonization with A. pomorum or L. plantarum or under germ free conditions) and a reduced larval viability in animals raised under both normal and low yeast conditions.

pHCl-2pTV-Cherry homozygous larvae -raised under low yeast conditions-

exhibit decreased mouth hook contractions per minute, a preference for zinc-supplemented food over time and a defective acidification of autophagic compartments in interstitial cells, and they are more translucent with less food in their intestines.

pHCl-2pTV-Cherry homozygous larvae -raised under low yeast conditions- exhibit reduced luminal acidity in the copper cell region of midgut, enlarged volumes of both the lumen of the copper cell region and the interstitial cells, which also exhibit a reduction in the complexity of their characteristic basal infoldings.

pHCl-2pTV-Cherry homozygous larvae exhibit increased gut bacterial titres following re-colonization with A. pomorum or L. plantarum.
