Detailed Description
Statement

Acn1/Acn27 animals show a distributed lethal phase, with most larvae dying during the third instar, while some die after pupariation.

Hemocytes isolated from Acn1/Acn27 third instar larvae appear indistinguishable from wild-type hemocytes in response to a treatment which induces apoptosis in wild-type hemocytes; the mutant hemocytes undergo nuclear condensation and chromatin fragmentation.

Autolysosomes are fewer and less abundant, while autophagosomes are larger and more abundant in the fat body of Acn1/Acn27 third instar larvae than in control larvae after starvation.

The severity of the notched wing phenotype seen in N264-39/+ flies is enhanced by Acn1/+.

Expression of TorTED.Scer\UAS under the control of Scer\GAL4Lsp2.PH cannot induce autolysosome formation in the fat body in a Acn1/Acn27 background.

Expression of PtenScer\UAS.cGb under the control of Scer\GAL4Lsp2.PH cannot induce autolysosome formation in the fat body in a Acn1/Acn27 background.

Comments

Acn+t4.2 and Acn4.2.T:Hsap\MYC each restore the viability of Acn1/Acn27 animals, and rescue the endocytosis and autophagy defects of Acn1/Acn27 larvae.

Wright.

The chromosome probably carries a second-site mutation, because the lethality of Acn1/Acn27 transheterozygotes, but not of Acn1 homozygotes, can be rescued by Acn+t4.2 or by Acn4.2.T:Hsap\MYC.
