Expression of tkvQ253D.Scer\UAS.cLa in the germline, under the control of Scer\GAL4nos.UTR.T:Hsim\VP16, does not generate any abnormalities in somatic development. There are little if any defects in germ cell migration and most primordial germ cells are properly aligned with the somatic germ cell precursors in stage 13 embryos. These mutants exhibit a substantial reduction in the number of germ cells on the coalesced gonad, apparently due to apoptosis.

Expression of tkvQ253D.Scer\UAS.cLa under the control of Scer\GAL4CY2 results in eggshells that lack dorsal appendages and have an operculum that is increased in size.

tkvQ253D.Scer\UAS.cLa; Scer\GAL4Ser.PGF animals raised at 25oC die as pupae. Those raised at 18oC survive to adulthood but have deformed, wrinkled wings.

Flies expressing tkvQ253D.Scer\UAS.cLa under the control of Scer\GAL4GMR.PF have rough, glassy eyes.

The number of cells in the lateral trunk anterior branch is not altered by expression of tkvQ253D.Scer\UAS.cLa under the control of Scer\GAL4btl.PS. There is an excess of dorsal branch cells in these embryos.

When tkvQ253D.Scer\UAS.cLa is driven by Scer\GAL4604 extravein tissue surrounds wing vein L5, which severely distorts and thickens the wing.

Embryos expressing tkvQ253D.Scer\UAS.cLa under the control of Scer\GAL469B show partial dorsalisation of their cuticles.

When tkvQ253D.Scer\UAS.cLa is driven by Scer\GAL4btl.PS tracheal cells are induced to migrate in the dorsal ventral axis. The gap between the most dorsal muscle precursors and the more ventral ones are missing.

When expressed via Scer\GAL469B, tracheal tree is modified such that only dorsoventral tracheal branches arise while longitudinal branches fail to form. When expressed via Scer\GAL4btl.PS, causes failure of the tracheal cells to migrate anteroposteriorly to form the dorsal trunk. The visceral branches and the lateral trunk usually form.

Scer\GAL4btl.PS-mediated expression diverts cells of the dorsal trunk and visceral branch primordia to the dorsal branch.

Produces ectopic vein tissue in the wing when expressed using Scer\GAL4C-580 or Scer\GAL4bs-1348.

The deformed, wrinkled wing phenotype seen in tkvQ253D.Scer\UAS.cLa; Scer\GAL4Ser.PGF flies is completely suppresessed by TolloScer\UAS.cKa or Tolloex.Scer\UAS.

The small eye phenotype of upd1unspecified/Df(1)os1A2 flies is partly suppressed by expression of tkvQ253D.Scer\UAS.cLa under the control of Scer\GAL4ey.PH. The rough eye phenotype caused by expression of tkvQ253D.Scer\UAS.cLa under the control of Scer\GAL4GMR.PF is strongly suppressed by glunspecified. The eye phenotype caused by expression of tkvQ253D.Scer\UAS.cLa under the control of Scer\GAL4GMR.PF is partially modified by Madunspecified.

The cuticle phenotype of embryos expressing tkvQ253D.Scer\UAS.cLa under the control of Scer\GAL469B is not altered if they are derived from homozygous shark1 female germ-line clones (and fertilised by a shark+ sperm). The dorsal closure defects of homozygous shark1 embryos derived from shark1 female germline clones are not rescued by expression of tkvQ253D.Scer\UAS.cLa under the control of Scer\GAL469B.

When tkvQ253D.Scer\UAS.cLa and armS10.Scer\UAS.T:Hsap\MYC are co-expressed using Scer\GAL4btl.PS, a partial restoration of the dorsal trunk is seen in the tracheal system.

When expressed via Scer\GAL4btl.PS in a vvlGA3 mutant background, no migration of tracheal cells occurs unless btl is also expressed, via btlhs.PM.

The ectopic vein phenotype seen when tkvQ253D.Scer\UAS.cLa expressed using Scer\GAL4C-580 or Scer\GAL4bs-1348 is reduced in combination with HLHmβScer\UAS.cdCa or phlDN.Scer\UAS.
