Lethality occurs during third instar larval or pupal stages. Mutants exhibit visible disc abnormalities. In tergites, clones with normal size and frequency are recovered. Clones cannot be recovered in the wing disc.
Brain, imaginal disc and blood cell neoplastic phenotype.
Brain is of normal size but abnormal shape. Wing and haltere discs rarely showing hyperplastic outgrowths, folding abnormal. Lymph gland retains lobular structure.
This allele was listed in the BDGP database as a lethal or sterile line during the period 1994-1999, but was discarded from the gene disruption project prior to the summary publication (FBrf0111489). Reasons for excluding lines from the collection described in FBrf0111489 include presence of more than one P insertion on the mutant chromosome, separation of lethality (or sterility) from the location of the insertion, and loss of lethality (or sterility) from the stock. The level of SmD3 transcript is strongly reduced in P{lacW}SmD3k13107 homozygotes compared to wild type, whereas Oda expression remains essentially unaffected. The level of SmD3 protein is lowered compared to wild type in P{lacW}SmD3k13107/+ larvae and strongly reduced in P{lacW}SmD3k13107 homozygous larvae. The P{lacW}SmD3k13107 insertion appears to affect only SmD3 function and not Oda function (despite being within an Oda intron), since the lethality of P{lacW}SmD3k13107 homozygotes is fully rescued by the P{SmD3.+t2.3} construct which contains the complete SmD3 gene. Two or three different shades of eye colour were observed, this phenotype is caused by the presence of a P{lacW} insert into another autosome cytological location 49B. P-element excision from cytological location 48E demonstrates the 48E insertion to be the cause of lethality.
