Itprug3/Itprwc703 adults show a "spread out" wing posture.

itgr[wu] flies (Itp-r83Awc703/Itp-r83Aug3) are resistant to starvation, but show a wild type response to oxidative stress. Females survive longer than males.

Itp-r83Awc703/Itp-r83Aug3 animals have wing posture, flight and flight physiology defects.

Itp-r83Awc703/+ 30-day-old adult males display impaired (but not abrogated) learning ability as they demonstrate only a weak decrease in courtship activity when paired with an mated unreceptive female but their short-term (60 min) memory is not impaired.

The spontaneous firing rate seen in dorsal-longitudinal muscles of Itp-r83Awc361/Itp-r83Awc703 adults is normal. These adults can be divided into fliers and non-fliers (assayed using a cylinder drop test). About 30% are non-fliers - exhibiting only brief rhythmic flight patterns that are usually not sustained beyond 5 seconds in response to stimulation with an air puff. In contrast, the non-fliers exhibit sustained flight patterns (>30seconds) in response to the same stimulus.

Itp-r83Awc703/Itp-r83Aug3 flies show a "down-turned" wing phenotype.

Itp-r83Awc703/Itp-r83Awc361 flies show a significant degree of flightlessness but show normal wing posture.

Indirect flight muscles of wild-type flies can express short bursts of electrical activity that are associated with cleaning behavior. Itp-r83Awc703/Itp-r83Aug3 flies show an increase in spontaneous firing from these muscles, which is unrelated to cleaning behavior. The indirect flight muscles of these mutants fail to produce firing patterns in response to an air puff, which occurs in wild-type muscles.

Maintenance of adaptation to a butanol source in Itp-r83Awc703 mutants is strongly defective, and recovery to 50% of preadaptation response occurs in less than one-third of the time taken in wild-type. The dominant phenotype of Itp-r83Awc703 is further accentuated by the presence of the recessive alleles Itp-r83Awc361 and Itp-r83Aug3. This suggests that the Itp-r83A dosage affects recovery from adaptation.

The learning defects (measured in a courtship behavior assay) observed in the PsnB3, PsnI2 or the Itp-r83Awc703 single heterozygote 30 day-old males are suppressed in the PsnB3/+;Itp-r83Awc703/+ but not in the PsnI2/+;Itp-r83Awc703/+ double heterozygotes. Combination with a single copy of Itp-r83A90B.0 can however restore the short-term memory defects characteristic for both PsnB3 and PsnI2 aged heterozygote males.

The viability of Itp-r83Awc703/Itp-r83Awc361 animals is unaffected by sl1/sl1 or sl2/sl2. It is also unaffected by Gα49B221c/+, but the resulting adults have abnormal wing posture, an enhanced incidence of flight defects and mild endogenous hyperactivity of spontaneous firing recorded from the dorsal longitudinal muscles. Wing posture and flight defect phenotypes are enhanced and viability is reduced if Plc21Ck31911/+ is also present, but partially suppressed by Gα49BScer\UAS.cRa; Scer\GAL4Ddc.PL and completely or largely suppressed by Ca-P60AKum170/+, even when Plc21Ck31911/+ is also present.

Complete rescue of the altered wing posture is seen in Itp-r83AScer\UAS.cVa; Scer\GAL4Ilp2.PR/+; Itp-r83Awc703/Itp-r83Aug3 animals. Flight ability is also rescued significantly in these flies. The delivery of an air puff stimulus to Itp-r83Awc703/Itp-r83Aug3 flies expressing Scer\GAL4Ilp2.PR-driven Itp-r83AScer\UAS.cVa leads to rhythmic generation of action potentials, but in most flies the air puff response lasts for a shorter period of time compared with the control flies.

Rescue of Itp-r83Awc703/Itp-r83Aug3 flies by expression of Itp-r83AScer\UAS.cVa under the control of Scer\GAL4hs.PB following a heat shock shows that expression during early pupal development is sufficient to rescue flight behavior and physiology, while expression after adult emergence is ineffective in rescuing flight.
