Expression of SRm160GE25979 under the control of Scer\GAL4da.PU results in lethality at 25[o]C. A few female escapers are seen, but no males. These females show no visible phenotypes.

Expression of SRm160GE25979 in the genital disc under the control of Scer\GAL4esg.PU yields mostly normal progeny at 25[o]C and both sexes are fertile. 10% of the males show abnormal genital structure, including rotated genitals or extra appendages or completely lacking external genitalia and analia. Sex combs and pigmentation are both normal. Animals with aberrant genital morphology have testes that are poorly developed and have failed to elongate, but these contain sperm of normal morphology.

Expression of SRm160GE25979 under the control of Scer\GAL4GMR.PU results in rough and shiny eyes, observable at 25[o]C with males more strongly affected. Loss of differentiation and complete disorganisation of the ommatidial array is seen compared to wild type controls. No sign of the pigment cell lattice is visible but the lens is present. Formation of the lamina is also abnormal in Scer\GAL4GMR.PU>SRm160GE25979 flies compared with wild type. Increased levels of apoptosis (activated caspases) are seen at 25[o]C compared to controls. A more severe phenotype is seen at 29[o]C: very few flies survive and those that do are females with glassy eyes with little pigment. Male lethality occurs during pupation.

SRm160G18603/SRm160GE25979 mutant flies are normal in all cuticular phenotypes and fertile.

SRm160GE25979 strongly enhances the female to male somatic sex transformation phenotype seen in DoaHD/DoaDem mutant flies. The male genital arch and "claspers" are seen in XX individuals.

One copy of Df(3L)H99 partially suppresses the eye roughness seen when SRm160GE25979 is expressed under the control of Scer\GAL4GMR.PU.

Expression of Diap1Scer\UAS.cUa suppresses the eye roughness seen when SRm160GE25979 is expressed under the control of Scer\GAL4GMR.PU.

One copy of DroncI24 slightly suppresses the eye roughness seen when SRm160GE25979 is expressed under the control of Scer\GAL4GMR.PU.

One copy of DroncI29 slightly suppresses the eye roughness seen when SRm160GE25979 is expressed under the control of Scer\GAL4GMR.PU.

One copy of Dcp-1k05606 slightly suppresses the eye roughness seen when SRm160GE25979 is expressed under the control of Scer\GAL4GMR.PU.

One copy of p535A-1-4 does not suppress the eye roughness seen when SRm160GE25979 is expressed under the control of Scer\GAL4GMR.PU.

One copy of p5311-1B-1 does not suppress the eye roughness seen when SRm160GE25979 is expressed under the control of Scer\GAL4GMR.PU.

One copy of DoaHD partially suppresses the lethality seen when SRm160GE25979 is expressed under the control of Scer\GAL4da.PU. The viable escapers do not display any cuticular phenotypes.

DoaHD/DoaDem does not suppress the lethality seen when SRm160GE25979 is expressed under the control of Scer\GAL4da.PU.

One copy of DoaDem partially suppresses the ommatidial disruption seen when SRm160GE25979 is expressed under the control of Scer\GAL4GMR.PU. The various cell types and layers are all identifiable albeit somewhat perturbed. Although differentiation pigment cells are observed and cone cells secrete a cornea of normal appearance, the pigment and photoreceptor cells fail to elongate to form a normal retina. There is a reduction in the levels of apoptosis (caspase activation).

DoaHD/DoaDem almost completely suppresses the smooth, glassy eye seen when SRm160GE25979 is expressed under the control of Scer\GAL4GMR.PU. The ommatidial disruption is partially suppressed: the various cell types and layers are all identifiable albeit somewhat perturbed. Although differentiation pigment cells are observed and cone cells secrete a cornea of normal appearance, the pigment and photoreceptor cells fail to elongate to form a normal retina. There is a reduction in the levels of apoptosis (caspase activation).

One copy of DoaHD enhances the loss of external genitalia phenotype seen in males expressing SRm160GE25979 under the control of Scer\GAL4esg.PU.

One copy of DoaDem enhances the loss of external genitalia phenotype seen in males expressing SRm160GE25979 under the control of Scer\GAL4esg.PU. Unlike in Scer\GAL4esg.PU>SRm160GE25979 mutant flies alone, a small proportion of females lack external genitals and anal plates.

Very few animals survive when SRm160GE25979 is expressed under the control of Scer\GAL4esg.PU in a DoaHD/DoaDem mutant background, but those that do survive completely lack genitalia and analia and die shortly following eclosion.
