In DoaDem/Doa01705b mutant flies, the leg segments distal to the first tarsus often fail to elongate. The femur and claws form normally. Defects are seen in both the mesothoracic and metathoracic tarsi.
The L5 wing vein in DoaRemγA/DoaDem mutant flies fails to attach to the anterior wing margin. Spurs are also seen in the posterior crossvein.
The ovaries of DoaE786/DoaDem mutant flies appear normal, despite asymmetry in the female genital sex combs.
In DoaDem/Doa198-14 mutants where the DoaDem allele is from the mother there are no survivors. When the Doa198-14 allele comes from the mother the flies are viable, but the postalar bristles are duplicated and the scutellar bristles are forked-like.
In DoaDem/Doa04743 mutants where the DoaDem allele is from the mother only rare pupae eclose and weak adults die quickly. The flies exhibit rough eyes, missing bristles and an incomplete L5 wing vein. When the Doa04743 allele comes from the mother there are no survivors.
Transheterozygotes with Doa01705b have rough eyes and duplicated scutellar bristles.
XX ; Doaunspecified/DoaDem adults have subtle intersexual characteristics in sex combs, pigmentation and genitalia. Female-specific vaginal bristles are missing or irregular in most cases. XX ; Doa7/DoaDem adults have subtle intersexual characteristics.

SRm160B103 strongly enhances the female to male somatic sex transformation phenotype seen in DoaHD/DoaDem mutant flies. The male genital arch and "claspers" are seen in XX individuals.
SRm160B103 produces a female to male somatic sex transformation phenotype in DoaHD/Doa7 mutant flies. The male genital arch and "claspers" are seen in XX individuals.
SRm160GE25979 strongly enhances the female to male somatic sex transformation phenotype seen in DoaHD/DoaDem mutant flies. The male genital arch and "claspers" are seen in XX individuals.
DoaHD/DoaDem does not suppress the lethality seen when SRm160GE25979 is expressed under the control of Scer\GAL4da.PU.
DoaHD/DoaDem almost completely suppresses the smooth, glassy eye seen when SRm160GE25979 is expressed under the control of Scer\GAL4GMR.PU. The ommatidial disruption is partially suppressed: the various cell types and layers are all identifiable albeit somewhat perturbed. Although differentiation pigment cells are observed and cone cells secrete a cornea of normal appearance, the pigment and photoreceptor cells fail to elongate to form a normal retina. There is a reduction in the levels of apoptosis (caspase activation).
One copy of DoaDem partially suppresses the ommatidial disruption seen when SRm160GE25979 is expressed under the control of Scer\GAL4GMR.PU. Although differentiation pigment cells are observed and cone cells secrete a cornea of normal appearance, the pigment and photoreceptor cells fail to elongate to form a normal retina. There is a reduction in the levels of apoptosis (caspase activation).
One copy of DoaDem enhances the loss of external genitalia phenotype seen in males expressing SRm160GE25979 under the control of Scer\GAL4esg.PU. Unlike in Scer\GAL4esg.PU>SRm160GE25979 mutant flies alone, a small proportion of females lack external genitals and anal plates.
Very few animals survive when SRm160GE25979 is expressed under the control of Scer\GAL4esg.PU in a DoaHD/DoaDem mutant background, but those that do survive completely lack genitalia and analia and die shortly following eclosion.
Expression of bamScer\UAS.P\T.T:Avic\GFP under the control of either Scer\GAL4bam.T:Hsim\VP16 or Scer\GAL4vas.2.6 promotes the differentiation of spermatogonia to spermatocytes in DoaDem/DoaHD testes.
DoaHD Ef1γGE29510/DoaDem flies show a number of defects, including ectopic wing venation, failure of either of the crossveins to attach (particularly the posterior crossvein in males), aberrantly formed crossveins, occasional eye roughness, curved wings and rarely, partial rotation of the female genitalia.
DoaDem in combination with E(gl)3Cunspecified results in flies with normal eyes, wings and bristles. No suppression of wa is seen in these flies.
Heterozygous traunspecified/+ adults have normal sexual differentiation. XX ; traunspecified/+ adults that are also carrying Doaunspecified/DoaDem are clearly intersexual with respect to the number or morphology of sex combs. XX; Doaunspecified traunspecified/DoaDem + adults have genitalia which have characteristics of both sexes, including male-like clasper bristles and anal pads. Pigmentation of abdominal tergites is intermediate to that of the normal male and female phenotypes. XY flies carrying traHsp83.PS develop phenotypically as nearly perfect females. This female differentiation is substantially reversed if the flies are also carrying Doaunspecified/DoaDem.
