Description

The R18D09 enhancer fragment is fused upstream of the Drosophila synthetic core promoter (DSCP) followed by the GAL4(DBD)::Zip- hemidriver (component of the 'Split GAL4' system, contains the GAL4 DNA-binding domain). A Hsp70 transcriptional terminator is present.

Elimination of molecular clock function (using cycΔ.UAS.Tag:MYC) in neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R18D09 and Hsap\RELAAD.R12G04 in adults does not affect the strength of the circadian rhythm of sleep-wake cycles, compared to controls.

Inhibition (using Hsap\KCNJ2UAS.EGFP) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R18D09 and Hsap\RELAAD.R12G04 significantly reduces the strength of the circadian rhythm of sleep-wake cycles, compared to controls; this is also the case when inhibition is restricted to the adult stage using a Gal80[ts] transgene.

Inhibition (using Hsap\KCNJ25XUAS.EGFP) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R18D09 and Hsap\RELAAD.VT057469 in third instar larvae does not significantly affect short-term associative appetitive olfactory memory following training with fructose, innate odor (n-amylacetate) preference or innate fructose preference, compared to controls.

Optogenetic activation (using Cnoc\ChR1Cs.IVS.20xUAS.Venus with 617 nm (red) light) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R18D09 and Hsap\RELAAD.R58E02 in adults results in a significant attraction to the illuminated area, no attraction is observed in controls.
