Nlg4KO10 mutant adults do not show adult locomotor activity defects.

Unlike in wild-type, the axon of L4 lamina neuron in Nlg4KO10 homozygous adults is not restricted to a single medulla column but often extends to neighboring columns.

Although the overall morphology of neuromuscular junctions (NMJs) in Nlg4KO10 as well as Nlg4KO10/Df(3R)ED6027 mutant third instar larvae appears unperturbed, the axon branches are significantly shortened and the number of boutons reduced. No defects in the number of presynaptic vesicles or T-bar morphology in Nlg4KO10 NMJ boutons, but the bouton area per active zone is increased as is the single (but not overall) postsynaptic density length. Nlg4KO10 mutants however show active zone structure defects: the presynaptic membrane is detached from the opposite postsynaptic membrane at several points and display invaginations. The area of subsynaptic reticulum per bouton is decreased.

Both Nlg4KO10 homozygous and Nlg4KO10/Df(3R)ED6027 larvae display small but significant decrease in the quantal content as well as the amplitude of evoked excitatory junction potentials (EJPs), but not the miniature EJPs (mEJPs). The frequency of mEJPs is dramatically increased in the mutants as compared with wild-type controls. Nlg4KO10 and Nlg4KO10/Df(3R)ED6027 mutant larvae also show reduced locomotion (quantified as distance crawled within 3-min time limit)

The frequency of L4 lamina neuron axons aberrantly invading adjacent medulla columns in adult Nlg4KO10,Nrx-1Δ83 double mutants is not significantly different from each of the single mutants.

The impaired columnar restriction of L4 lamina neuron axons (which frequently invade adjacent medulla columns) observed in adult flies expressing EphrinHMS01289 under the control of Scer\GAL4GMR31C06 is not worsened further by combination with Nlg4KO10 in homozygous state - either of which on their own also causes defects in columnar restriction of the L4 axons.

Although none of the larvae heterozygous for any of the following alleles: Nlg4KO10, tkv7, witA12 or Mad8-2 display any significant difference in the number or size of boutons on neuromuscular junctions compared to controls, double heterozygotes for Nlg4KO10 and tkv7 or witA12 or Mad8-2 all show reduced bouton number and increased bouton size.

The synaptic overgrowth phenotype (increased number of boutons at neuromuscular junctions) characteristic for third instar larvae homozygous for Dadj1E4 is completely suppressed by Nlg4KO10 homozygosity and the resulting number of boutons per NMJ is comparable to Nlg4KO10 single mutants and significantly lower than in wild-type controls.

The impaired columnar restriction of L4 lamina neuron axons (which frequently invade adjacent medulla columns) observed in Nlg4KO10 mutant adult flies is rescued by expression of Nlg4Scer\UAS.cLa under the control of Scer\GAL4oc.1.6 in the mutant background.

The decreased number but increased size of boutons on neuromuscular junctions in Nlg4KO10 mutant third instar larvae is fully rescued by expression of Nlg4Scer\UAS.cZa if driven by either Scer\GAL4elav.PU or Scer\GAL4RapGAP1-OK6 but not Scer\GAL4C57. Similarly, the functional neurotransmission defects (reduced quantal content, decreased amplitude of evoked excitatory junction potentials) along with the larval locomotor deficit are rescued by Scer\GAL4elav.PU-driven expression of Nlg4Scer\UAS.cZa but not if it is controlled by Scer\GAL4C57.
