A 30 minute heat shock several hours before the beginning of dorsal closure in embryos carrying JraAsp.hs.sev results in less than 10% of embryos having defects. Those that do have very small anterior holes in the cuticle.
Flies have rough eyes with ommatidia that often contain extra R7 cells. Most ommatidia with the normal complement of photoreceptors are symmetrical R3/R3-type clusters and/or are mis-rotated. The phenotype resembles that of fzScer\UAS.cAa driven by Scer\GAL4hs.2sev. The eye disc reveals early polarity defects in the irregular arrangement and rotation angles of the clusters. Mosaic analysis reveals that JraAsp.hs.sev induces the R3 fate, as does over-activation of the fz pathway.
Expression causes transformation of cone cell precursors to photoreceptor neurons.
Eye surface is irregular and the ommatidia often contain ectopic, extra R7-like photoreceptors. The extra R7 cells differentiate from cone cell precursors. These R7 cells are functional as determined by behavioral tests in a sev14 background.

Embryos derived from DokPG155 germ-line clones that carry one copy of the JraAsp.hs.sev transgene exhibit a less severe dorsal closure effect, both in terms of expressivity and penetrance, than DokPG155/Y embryos. The transgene-carrying embryos die at late embryonic stage and exhibit small anterior holes in the cuticle.
The dorsal open phenotype seen in homozygous Cka05836 embryos derived from females carrying homozygous Cka05836 germ-line clones is substantially rescued by expression of JraAsp.hs.sev using heat shock.
The severe dorsal open phenotype of slpr921/Y embryos is significantly rescued by expression of JraAsp.hs.sev (using a 30 minute heat shock several hours before the beginning of dorsal closure).
The dorsal open phenotype seen in some Btk29Ak00206 Src42Amyri/Btk29Ak05610 Src42Amyri embryos is partially rescued by JraAsp.hs.sev.
Reduction of pnt dosage suppresses the supernumerary photoreceptor phenotype but has no effect on the tissue polarity defect.
JraAsp.hs.sev cannot rescue the eye phenotype of pnt mutants, though pnt mutants can rescue the eye phenotype of JraAsp.hs.sev. phyl mutants also strongly suppress the eye phenotype of JraAsp.hs.sev.
