Homozygous cicfet-U6 intestinal stem cell somatic clones (generated by MARCM) are larger than control clones and contain more cells.
Number of mitotic cells per midgut is significantly increased in cicfet-U6/cicfet-E11 transheterozygous adults.
Eggs derived from egg chambers containing complete homozygous follicle cell clones have a dorsalised eggshell.
Eggs from mosaic homozygous cicfet-U6 females exhibit ventral and lateral foci of ectopic appendage-like materail. Analysis of homozygous cicfet-U6 follicle cell clones confirms that these foci are associated with ventral and lateral clones of cicfet-U6 mutant clones. This ectopic appendage material is restricted to the anterior half of the eggshell, while posterior clones exhibit no detectable phenotype. Interestingly, ectopic appendage material at the extreme anterior of the eggshell often appears as appendage-like projections, suggesting that only cicfet-U6 mutant follicle cells in the corresponding region of the columnar follicular epithelium can form an appendage-producing epithelial tube.
With the exception of those at the dorsal midline, cicfet-U6 mutant follicle cell clones undergo apical constriction reminiscent of that observed in the endogenous appendage-producing populations. In addition, the follicle cell nuclei appear more closely packed in both the endogenous appendage roof primordia and in regions of anterior cicfet-U6 mutant cells, presumably as a consequence of this apical constriction.
Approximately 12.9% of cicfet-E11/cicfet-U6 females exhibit a weak eggshell phenotype. The dorsal midline is expanded and the appendages are broader and positioned laterally with a trace of ventral collar material. The dorsalmost region remains free of appendage material.
Approximately 19.3% of cicfet-U6/cicfet-E11 females exhibit a moderate eggshell phenotype. The dorsal midline is expanded and the appendages are broader and positioned laterally and exhibit a ventral collar of ectopic appendage-like material. The dorsalmost region remains free of appendage material.
Approximately 30.0% of cicfet-U6/cicfet-E11 exhibit a moderate-severe eggshell phenotype. The dorsal midline is expanded and the appendages are broader and positioned laterally and exhibit a pronounced ventral collar of ectopic appendage-like material. The dorsalmost region remains free of appendage material.
Approximately 14.1% of cicfet-U6/cicBA53 exhibit a moderate-severe eggshell phenotype. The dorsal midline is expanded and the appendages are broader and positioned laterally and exhibit a pronounced ventral collar of ectopic appendage-like material. The dorsalmost region remains free of appendage material.
Approximately 85.9% of cicfet-U6/cicBA53 exhibit a severe eggshell phenotype. The dorsal midline is expanded and the appendages are broader and positioned laterally and exhibit a pronounced ventral collar of ectopic appendage-like material, along with small anterior projections.
Approximately 32.8% of cicfet-U6/cicfet-T6 females exhibit a weak-moderate eggshell phenotype. The dorsal midline is expanded and the appendages are broader and positioned laterally and exhibit a ventral collar of ectopic appendage-like material. The dorsalmost region remains free of appendage material.
Approximately 20.9% of cicfet-U6/cicfet-T6 exhibit a moderate-severe eggshell phenotype. The dorsal midline is expanded and the appendages are broader and positioned laterally and exhibit a pronounced ventral collar of ectopic appendage-like material. The dorsalmost region remains free of appendage material.
Approximately 1.7% of cicfet-U6/cicfet-T6 exhibit a severe eggshell phenotype. The dorsal midline is expanded and the appendages are broader and positioned laterally and exhibit a pronounced ventral collar of ectopic appendage-like material, along with small anterior projections.
Eggs derived from cicfet-U6/cicfet-E11 females are dorsalised. The eggs are shorter than normal and have broader dorsal appendages that are shifted laterally, with appendage base material encircling the entire anterior circumference of the egg. Embryos derived from cicfet-U6/cicfet-E11 females fail to hatch and are dorsalised (the secreted cuticle consists entirely of dorsal epidermis). cicfet-U6/cicfet-E11 adults have smaller wings than normal, with vein thickening and blistering. 22% of eggs produced by females with homozygous germline clones are dorsalised (78% are wild type), but 100% of the embryos derived from these eggs fail to hatch (no cellularisation is seen in these embryos). 34% of eggs derived from females carrying homozygous follicle cell clones are dorsalised (66% are wild type). 54% of the embryos derived from these females fail to hatch (of these, 10% are locally dorsalised along the anterior-posterior axis and 1% are completely dorsalised), and 46% hatch. Dorsal or lateral follicle cell clones do not result in defects in the dorso-ventral patterning of the embryo, while ventral follicle cell clones produce local dorsalisation in the embryo. cic1/cicfet-U6 females produce embryos with a tor gain-of-function phenotype.

Approximately 0.5% of cicfet-T6/mirr6D1 cicfet-U6 females exhibit a weak eggshell phenotype. The dorsal midline is expanded and the appendages are broader and positioned laterally with a trace of ventral collar material. Approximately 42.6% of cicfet-U6/mirr6D1 cicfet-T6 females exhibit a moderate eggshell phenotype. The dorsal midline is expanded and the appendages are broader and positioned laterally and exhibit a ventral collar of ectopic appendage-like material. The dorsalmost region remains free of appendage material. Approximately 42.1% of cicfet-U6/mirr6D1 cicfet-T6 females exhibit a moderate-severe eggshell phenotype. The dorsal midline is expanded and the appendages are broader and positioned laterally and exhibit a thick ventral collar of ectopic appendage-like material. The dorsalmost region remains free of appendage material. None of the cicfet-U6/mirr6D1 cicfet-T6 females exhibit a severe eggshell phenotype (compared to 1.7% in the heterozygote). There is an increase in the weak phenotype, and a decrease in the severe phenotype compared to the cicfet-T6/cicfet-U6 mutant, indicating that mirr6D1 suppresses the cicfet-T6/cicfet-U6 phenotype.
Approximately 34.4% of cicfet-E11/mirr6D1 cicfet-U6 females exhibit a weak eggshell phenotype. Approximately 37.4% of cicfet-U6/mirr6D1 cicfet-E11 females exhibit a weak-moderate eggshell phenotype. Approximately 37.9% of cicfet-U6/mirr6D1 cicfet-E11 females exhibit a moderate-severe eggshell phenotype. The dorsal midline is expanded and the appendages are broader and positioned laterally and exhibit a thick ventral collar of ectopic appendage-like material. The dorsalmost region remains free of appendage material. There is an increase in the weak phenotype, and a decrease in the severe phenotype compared to the cicfet-E11/cicfet-U6 mutant, indicating that mirr6D1 suppresses the cicfet-E11/cicfet-U6 phenotype.

Stage 12 grk3/grk2B6 ; cicfet-U6/cicfet-T6 double mutant egg chambers lack dorsal appendages (characteristic of loss of grk), but retain a pronounced collar of appendage material, indicating that loss of grk cannot suppress the cic phenotype.
rho6 mutant clones have no effect on the ectopic appendage-producing fates in cicfet-U6/cicfet-T6 mutant egg chambers, indicating that loss of rho function cannot suppress the cic phenotype.
Selected as: a mutant that shows extra wing veins or blisters over cicfet-E11.
