Expression of NPFUAS.cWa under the control of Scer\GAL4NPF.1 in males does not affect courtship towards males and significantly increases aggressive lunging towards males, compared to controls.

The expression of NPFUAS.cWa under the control of Scer\GAL4NP1624 leads to virgin females showing an increase in the number of germline stem cells in the germarium, as compared to controls.

NPFsk1/Df(3R)ED10642 transheterozygous females fail to show the expected mating-induced increase in germline stem cells in the germarium, as compared to controls; the expression of NPFUAS.cWa under the control of Scer\GAL4Tk.PS rescues the germline stem cell number in mated females, but also leads to an abnormal increase in germline stem cells in virgin females.

Over-expression of NPFScer\UAS.cWa in Scer\GAL4NPF.1 neurons does not significantly affect total sleep amount compared to genetic controls.

When npfScer\UAS.cWa is driven by Scer\GAL4386Y or Scer\GAL4npf.1 mutant animals are highly sensitive to alcohol. When npfScer\UAS.cWa is driven by Scer\GAL4npf.1 mutant animals show a normal response to ethyl ether exposure.

Larvae expressing npfScer\UAS.cWa under the control of Scer\GAL4386Y and control larvae develop similarly during the feeding phase and have similar body sizes at 98 hours after egg laying. The path length of larvae expressing npfScer\UAS.cWa under the control of Scer\GAL4386Y crawling on water-agarose paste is similar to that of wild-type larvae. Older third instar larvae (100 hours after egg laying) expressing npfScer\UAS.cWa under the control of Scer\GAL4386Y which are placed on a glucose-agar plate coated with a thin layer of yeast paste browse evenly across the assay plate (the plate is examined after 45 minutes). This behaviour is not typical of older wild-type larvae, which tend to migrate to the periphery of the plate and form clumps of larvae at the periphery, but is instead typical of young wild-type third instar larvae. Older third instar larvae (100 hours after egg laying) expressing npfScer\UAS.cWa under the control of Scer\GAL4386Y placed on an apple juice-agar plate show no social interaction and completely fail to burrow into the agar, even after 1 hour. This is in contrast to control larvae, which show social burrowing and penetrate the agar in less than 30 minutes. Larvae expressing npfScer\UAS.cWa under the control of Scer\GAL4386Y do not stop feeding until 12-24 hours later than controls and the mutant larvae are delayed in forming puparia by about the same amount of time. In the absence of food, well-fed early third instar larvae expressing npfScer\UAS.cWa under the control of Scer\GAL4386Y and control larvae develop into puparia at the same rate. Non-feeding (120 hours after egg laying) larvae expressing npfScer\UAS.cWa under the control of Scer\GAL4386Y show neither food aversion nor social burrowing when placed on an apple juice-agar plate. Most of these larvae form puparia on the moist food-containing agar surface, in contrast to control larvae, which mostly form puparia on the dry plastic surface of the plate.

Reference

Co-expression of NPFScer\UAS.cWa does not significantly alter the decreases in sleep amount seen in Scer\GAL4NPF.1>TrpA1Scer\UAS.cUa flies at 29[o]C; total sleep in flies with over-expression is less than that seen in controls.
