dj-1βΔ93 mutant flies show elevated response across a range of spatiotemporal frequencies of contrast-reversing sine-wave gratings as compared to responses from control flies.

No rhabdomere loss is observed in dj-1βΔ93 mutant flies.

dj-1βΔ93/dj-1βΔ93 mutants display normal eye development and ultrastructure, and normal L4 wing vein morphology.

dj-1βΔ93/dj-1βΔ93 mutants do not display any significant decrease in fertility, but have a reduced lifespan and are increasingly climbing defective with age, as compared to controls.

dj-1βΔ93 mutant flies are are hyper-sensitive to oxidative stress in the form of paraquat exposure.

dj-1βΔ93 flies show no striking external phenotypes. dj-1βΔ93 flies show a much higher sensitivity to the oxidative agent paraquat compared to wild-type flies.

DJ-1αΔ72/+ dj-1βΔ93/dj-1βΔ93 significantly suppresses the small rough eye phenotype in RetMEN2A.GMR flies. dj-1βΔ93/+ or DJ-1αΔ72/DJ-1αΔ72 dj-1βΔ93/+ significantly suppresses the small rough eye phenotype in RetMEN2B.GMR flies. dj-1βΔ93/+ significantly suppresses the increase in ommatidia size in RetMEN2B.GMR flies.

dj-1βΔ93/dj-1βΔ93 does not suppress the eye phenotypes (including increased eye size) in flies with expression of Pi3K92EUAS.Tag:MYC driven by Scer\GAL4GMR.PU at 30[o]C. dj-1βΔ93/dj-1βΔ93 does not enhance eye and ommatidial size phenotypes in flies with expression of Akt1UAS.cUa driven by Scer\GAL4GMR.PU. dj-1βΔ93/+ does not suppress increases in eye size in flies with expression of Pi3K92EUAS.Tag:MYC,Tag:PM(hKRAS) driven by Scer\GAL4GMR.PU.

dj-1βΔ93/+ or dj-1βΔ93/dj-1βΔ93 does not suppress the decreases in eye size seen in flies with expression of PtenUAS.cKa driven by Scer\GAL4GMR.PU.

dj-1βΔ93/+ DJ-1αΔ72/+ suppresses eye phenotypes (including increase in R cell number per ommatidium) in Ras85DG12V.sev flies. dj-1βΔ93/+ does not alter eye phenotypes in flies with expression of rlSem.cUa.UAS driven by Scer\GAL4GMR.PU.

dj-1βΔ93/dj-1βΔ93 significantly enhances eye and L4 wing vein phenotypes in rl1/rl1 flies.

DJ-1αΔ72/DJ-1αΔ72, dj-1βΔ93/dj-1βΔ93 double mutant flies have a reduced lifespan; display decreased spontaneous movement and climbing activity over time; have reduced fertility in both males and females; but have normal mitochondrial morphology, as compared with controls. Sperm from DJ-1αΔ72/DJ-1αΔ72, dj-1βΔ93/dj-1βΔ93 males appear morphologically normal but are immotile, have abnormal vacuoles in the Nebenkern, irregularities of mitochondrial derivatives, and misoriented and asynchronous progression of the investment cones during spermatogenesis. Mitochondria in DJ-1αΔ72/DJ-1αΔ72, dj-1βΔ93/dj-1βΔ93 double mutant flies show no difference in generating and maintaining cross-membrane potential, but do show a decline in mitochondrial function as the flies age, as measured by comparing the respiratory control ratio in 3 day- and 30 day-old whole flies.

Expression of parkScer\UAS.cGa or Pink1Scer\UAS.cUa under the control of Scer\GAL4da.PU results in lethality in the DJ-1αΔ72/DJ-1αΔ72, dj-1βΔ93/dj-1βΔ93 double mutant background.

DJ-1αΔ72; dj-1βΔ93 flies are viable, fertile and have no striking external phenotypes. There are no differences in the number of DA neurons in the fly brain between young (1 day) or aged (30 days) flies. DJ-1αΔ72; dj-1βΔ93 flies show a higher sensitivity to the oxidative agents paraquat and H2O2 than wild-type flies. However, the sensitivity of these double mutants to paraquat is not significantly higher than in dj-1βΔ93 single mutants. The double mutants do not show an increase in sensitivity to chemical agents that induce the unfolded protein response or that impair proteasome function. Expression of the either the dj-1βScer\UAS.cMa transgene or the DJ-1αScer\UAS.cMa transgene, under the control of Scer\GAL4αTub84B.PL, rescues the sensitivity of dj-1βΔ93; DJ-1αΔ72 mutants to paraquat.

DJ-1αΔ72; dj-1βΔ93 flies show a higher sensitivity to the oxidative agents paraquat, H2O2 and rotenone than wild-type flies.

Expression of Hsap\PARK7Scer\UAS.cMa under the control of Scer\GAL4da.G32 partially rescues the paraquat sensitivity of dj-1βΔ93 mutant flies, as compared to rescue by dj-1βScer\UAS.cMa expressed using the same driver.

Complementation and Rescue Data
Comments

Expression of dj-1βScer\UAS.cMa under the control of Scer\GAL4da.G32 rescues the paraquat sensitivity of dj-1βΔ93 flies.

Expression of dj-1βC45A.Scer\UAS or dj-1βC45D.Scer\UAS under the control of Scer\GAL4da.G32 rescues the paraquat and hydrogen peroxide sensitivity of dj-1βΔ93 flies.

Expression of dj-1βC104A.Scer\UAS, dj-1βC104D.Scer\UAS or dj-1βC45A.C104A.Scer\UAS under the control of Scer\GAL4da.G32 fails to rescue the paraquat or hydrogen peroxide sensitivity of dj-1βΔ93 flies.
