Description

An αTub67C promoter drives expression of amino acids 1-727 of the mira coding region, tagged with mGFP6. The expressed mira is lacking the C-terminal 103 amino acids compared to the full-length protein.

Expression of miraΔ103.αTub67C.T:Avic\GFP-m6 results in embryos that have an anterior abdomen in place of the head and thorax with 100% penetrance. These embryos also have ectopic anterior pole cells.

Females that express miraΔ103.αTub67C.T:Avic\GFP-m6 in a exuVL/exu4 background produce embryos with an exu phenotype, in which the thorax and abdomen develop normally, but the head is absent.

Embryos from females expressing miraΔ103.αTub67C.T:Avic\GFP-m6 in a swa1/swa3 background show the swa1/swa3 phenotype, in which the embryonic head is reduced but not lost.

Embryos that express miraΔ103.αTub67C.T:Avic\GFP-m6 and Sry-β::Sry-δDB56 in a Sry-δ14/Df(3R)X3F background show that same phenotype as embryos that express miraΔ103.αTub67C.T:Avic\GFP-m6 in a wild-type background.

Embryos that express both miraΔ103.αTub67C.T:Avic\GFP-m6 and stauΔloop2.αTub67C.T:Hsap\MYC in a stau2 background lack a posterior abdomen and form an anterior abdomen with complete reverse polarity.

Embryos that express miraΔ103.αTub67C.T:Avic\GFP-m6 in a mago1/Df(2R)F36 background have no posterior abdomen and have an anterior abdomen of reversed polarity. Embryos that express miraΔ103.αTub67C.T:Avic\GFP-m6 in a Tm1unspecified background show an identical reversed polarity phenotype.
