Expression of Zzzz\E4orf4Scer\UAS.cPa under the control of Scer\GAL4GMR.PF at 29[o]C induces a small and rough eye phenotype. The severity of this phenotype is reduced at lower temperatures and is more dramatic in males than in females. At 29[o]C eye size is reduced to 48% of controls in males and 68% of controls in females.

Expression of Zzzz\E4orf4Scer\UAS.cPa under the control of Scer\GAL4en.PU causes a variety of wing defects, including loss of the anterior cross-vein, loss of the posterior cross-vein, partial elimination of the L5 vein, partial elimination of the posterior part of the wing blade, and almost complete elimination of the posterior part of the wing. The severity and penetrance of this phenotype is increased at higher temperatures. The wings of flies raised at 18[o]C are larger than controls, but this effect is not seen at higher temperatures. Dose-dependent loss of the alulae bristles is also observed.

Flies expressing Zzzz\E4orf4Scer\UAS.cPa under the control of Scer\GAL4GMR.PF are fully viable at either 29[o]C or 18[o]C.

100% of third instar larval wing discs expressing Zzzz\E4orf4Scer\UAS.cPa under the control of Scer\GAL4en.PU at 29[o]C contain a cluster of dead cells (expressing caspase).

Expression of thScer\UAS.T:Hsap\MYC.cBa partially rescues the eye phenotypes seen when Zzzz\E4orf4Scer\UAS.cPa is expressed under the control of Scer\GAL4GMR.PF at 29[o]C. Eye size is restored to 75% and 87% of controls in males and females respectively. Eyes still appear rough and shape is disrupted.

Expression of BacA\p35Scer\UAS.P\T.cBa partially rescues the eye phenotypes seen when Zzzz\E4orf4Scer\UAS.cPa is expressed under the control of Scer\GAL4GMR.PF at 24[o]C. In females the resulting eye size is comparable to wild type, whereas males eyes are ~85% the size of those in wild type flies. Eyes still appear rough and shape remains disrupted.

Expression of Zzzz\E4orf4Scer\UAS.cPa suppresses the semi-viability seen when WScer\UAS.cUa is expressed under the control of Scer\GAL4GMR.PF at 18[o]C. 95% of pupae eclose (compared to 69%). The reduction in eye size is also partially suppressed.

Expression of Zzzz\E4orf4Scer\UAS.cPa partially suppresses the semi-lethality seen when rprScer\UAS.C is expressed under the control of Scer\GAL4GMR.PF at 18[o]C. 78% of pupae eclose (compared to 32.5%). The reduction in eye size is also partially suppressed.

Expression of Zzzz\E4orf4Scer\UAS.cPa partially suppresses the semi-lethality seen when rprScer\UAS.cZa is expressed under the control of Scer\GAL4GMR.PF at 18[o]C. 27.4% of pupae eclose (compared to 0.5%). The reduction in eye size is also partially suppressed.

Expression of Zzzz\E4orf4Scer\UAS.cPa suppresses the semi-lethality seen when grimScer\UAS.cUa is expressed under the control of Scer\GAL4GMR.PF at 18[o]C. 78% of pupae eclose (compared to 0.01%). The reduction in eye size is also partially suppressed.

Expression of Zzzz\E4orf4Scer\UAS.cPa suppresses the lethality seen when egrScer\UAS.cIa is expressed under the control of Scer\GAL4GMR.PF; 99% of adults eclose at 18[o]C, 100% at 24[o]C and 94% at 29[o]C.

Expression of Zzzz\E4orf4Scer\UAS.cPa partially suppresses the reduction in eye size seen when egrScer\UAS.cIa is expressed under the control of Scer\GAL4GMR.PF at 18[o]C. Eye size is increased to 39% in females (from 13%) and 53% in males (from 26%).

Expression of Zzzz\E4orf4Scer\UAS.cPa partially rescues the eye phenotypes seen when hepAct.Scer\UAS is expressed under the control of Scer\GAL4GMR.PF at 18[o]C. Eye size is restored to 23% and 31% of controls in females and males respectively (from 19% in each).

Expression of Zzzz\E4orf4Scer\UAS.cPa partially rescues the eye phenotypes seen when hepCA.Scer\UAS is expressed under the control of Scer\GAL4GMR.PF at 18[o]C. Eye size is restored to 25% and 31% of controls in females and males respectively (from 17% and 18%).

Expression of Zzzz\E4orf4Scer\UAS.cPa has little effect on the reduced eclosion rates seen when hepCA.Scer\UAS is expressed under the control of Scer\GAL4GMR.PF at 18[o]C. However at 24[o]C the reduced eclosion rate is partially suppressed, with 89% of flies eclosing (compared to 0.75% when hepCA.Scer\UAS is expressed alone).

One copy of egr1 enhances the reduction in eye size seen in both males and females when Zzzz\E4orf4Scer\UAS.cPa is expressed under the control of Scer\GAL4GMR.PF at either 24[o]C or 29[o]C. An enhancement is also seen in females at 18[o]C, but no difference is observed in males.

Expression of pucScer\UAS.cMa enhances the reduction in eye size seen in both males and females when Zzzz\E4orf4Scer\UAS.cPa is expressed under the control of Scer\GAL4GMR.PF at any of 18[o]C, 24[o]C or 29[o]C.

One copy of twsaar-1 partially suppresses the reduction in eye size seen when Zzzz\E4orf4Scer\UAS.cPa is expressed under the control of Scer\GAL4GMR.PF. Eyes are 83% of wild type size in females and 74% in males.

One copy of tws60 partially suppresses the reduction in eye size seen when Zzzz\E4orf4Scer\UAS.cPa is expressed under the control of Scer\GAL4GMR.PF. Eyes are of wild type size in females, but only 82% in males.

One copy each of twsaar-1 and tws60 completely suppresses the reduction in eye size seen when Zzzz\E4orf4Scer\UAS.cPa is expressed under the control of Scer\GAL4GMR.PF.
