Stripjam/Stripjam, Stripjam/Stripdogi, Stripjam/Df(3L)tc-1 and Stripjam/Df(3L)HR232 mutants exhibit defective L3 male : small gonads with supernumerary strongly DAPI-positive cells throughout, whereas wild-type and Stripjam/+ control gonads exhibit brighter DAPI-stained DNA in the apical region of the tissue, where the GSCs, gonialblasts, TA spermatogonia, CySCs and CyCs reside.

Further analysis of Stripjam/Stripjam L3 male gonads uncovers additional germline and somatic differentiation defects: the gonad is mainly occupied by small individualized germline cells containing small dot-like spectrosomes, rather than branched fusomes; stem cells (marked by Esg), spermatogonia (marked by Bam) are present throughout the gonad, instead of only being present at the tip/close to the tip of the gonad; early cyst cells can be found at the surface of the gonads, rather than interspersed between the germline cells; differentiated cysts and spermatocytes (marked by Topi) are observed only rarely; the germline lineage is less well enclosed by the somatic cell partners (marked by Arm); most somatic cells exhibit have disrupted morphology and disrupted nuclei morphology, and express markers for both early/stem cyst cell (Zfh1 or Tj) and differentiated cyst cell (Eya) fate. There are no obvious hub defects.

Individualized germline cells overproliferate (i.e. BrdU- or pH3-positive) and these dividing cells are present throughout the gonad, instead of only being present at the tip/close to the tip of the gonad. Apoptotic cells (i.e. TUNEL- or cleaved Caspase-3-positive) are also present throughout the gonad, while being negligible in controls.
