The regulatory N-terminal domain/NTD binds calcium in the mitochondrial intermembrane space and regulates the antiporter activity of the transmembrane domain/TMD. In absence of calcium, the apo form of the N-terminal domain is intrinsically disordered and binds to the transmembrane domain, inhibiting the transporter activity. Binding of calcium leads to a major conformational change and abolishes the interaction with the transmembrane domain and the inhibition of the transporter activity.
The C-terminal mitochondrial carrier domain/transmembrane domain/TMD bears the transmembrane transporter activity.
Linker region/H9 could directly block the transport of substrates across the transporter.

The testis specificity index was calculated from modENCODE tissue expression data by Vedelek et al., 2018 to indicate the degree of testis enrichment compared to other tissues. Scores range from -2.52 (underrepresented) to 5.2 (very high testis bias).
All three isoforms of SCaMC are expressed in the corpus allatum in the ring gland in larvae as well as in most larval tissues. Isoform-specific expression patterns are observed. SCaMC-RC is expressed at a 10X higher level in intestines and fat body than in the corpus allatum. All isoforms are expressed in adult testes and ovaries.
