Cas9(R691A) is an engineered RNA-guided nuclease derived from the naturally occurring Cas9 endonuclease from Streptococcus pyogenes. It contains a R691A mutation, resulting in a protein that retains the high on-target activity of Cas9 while reducing off-target editing (PMID:30082871).
