Expression of Src64BUY1332 in the posterior compartment of the wing imaginal disc, under the control of Scer\GAL4hh-Gal4 results in epithelial extrusion of cells moving to distant areas from the posterior compartment and activation of the apoptosome component Nc (and a subsequent increase in apoptosis).

Expression of Src64BUY1332 under Scer\GAL4sd-SG29.1 control at 25[o]C induces massive apoptosis. At 18[o]C, these tissues do not contain apoptotic cells.

Expression of Src64BUY1332 under the control of Scer\GAL4hh-Gal4 decreases the size of the posterior imaginal wing disc blade domain, relative to the total blade area, to 19.4% from 34.6% in controls.

Expression of Src64BUY1332 under the control of Scer\GAL4ap-md544 results in pupal lethality.

Larval wing discs expressing Src64BUY1332 in the dorsal compartment of the wing disc under the control of Scer\GAL4ap-md544 are grossly overgrown and exhibit elevated levels of apoptosis, extending beyond the dorsal compartment.

When expression is driven by Scer\GAL47B, Src64BUY1332 shows a mushroom body Class III phenotype; axonal growth and guidance defects. The beta lobes are abortive and the alpha lobes enlarged, particularly at the basal end. Phenotype is similar though more dramatic - all axons stop at the peduncle - when Src64BUY1332 is driven by Scer\GAL4OK107.

Larvae with expression of Src64BUY1332 driven in muscles by Scer\GAL4C57 show significant decreases in mEPSP (decreased quantal size) but do not show significant compensatory homeostatic increases in quantal content (as seen in GluRIIASP16/GluRIIASP16 mutants alone) at the NMJ; when expression is driven neuronally by Scer\GAL4elav-C155 the significant increase in quantal content is still seen.

Blocking JNK signalling, through expression of dominant-negative bskDN.Scer\UAS, significantly abolishes apoptosis and restores growth in imaginal wing discs expressing Src64BUY1332 under the control of Scer\GAL4sd-SG29.1. However, bskDN.Scer\UAS expression does not appear to prevent loss of epithelial integrity as cross-sections through these epithelia show the presence of dividing nuclei on the basal surface.

Co-expression of cpadsRNA.Scer\UAS.cFa and Src64BUY1332 under the control of Scer\GAL4nub-AC-62, results in massive apoptosis.

Co-expression of cpaScer\UAS.N.T:Ivir\HA1 and cpbScer\UAS.cWa appears to significantly suppress apoptosis and to restore tissue growth in imaginal wing discs expressing Src64BUY1332 under the control of Scer\GAL4sd-SG29.1.

Removal of one copy of shg, through a shgk03401 heterozygous background, restores the size of the posterior imaginal wing disc blade to 28.3% in flies expressing Src64BUY1332 under the control of Scer\GAL4hh-Gal4.

Removal of one copy of Btk29A, through a Btk29Ak00206 heterozygous background, significantly abolishes apoptosis and restores tissue growth in flies expressing Src64BUY1332 under the control of Scer\GAL4sd-SG29.1. However, this does not trigger the presence of mitotic nuclei on the basal cell surface.

Removal of one copy of p120ctn, through a p120ctn308 heterozygous background, significantly abolishes apoptosis and restores tissue growth in flies expressing Src64BUY1332 under the control of Scer\GAL4sd-SG29.1. However, this does not trigger the presence of mitotic nuclei on the basal cell surface.

Removal of one copy of Rho1, through a Rho11B heterozygous background, significantly abolishes apoptosis and restores tissue growth in flies expressing Src64BUY1332 under the control of Scer\GAL4sd-SG29.1. However, this does not trigger the presence of mitotic nuclei on the basal cell surface.

Apoptotic levels are not significantly different in tissues expressing cpadsRNA.Scer\UAS.cFa under the control of Scer\GAL4sd-SG29.1 that are either wild-type or heterozygous for Btk29Ak00206 or p120ctn308. While removing one copy of Rho11B enhances apoptosis in cpa-depleted tissues.

Expression of PRL-1Scer\UAS.cPa partially suppresses the pupal lethality seen when Src64BUY1332 is expressed in the dorsal compartment of the wing disc under the control of Scer\GAL4ap-md544, enabling 45% of the expected adults to eclose. The overgrowth and apoptosis seen in the larval wing discs are also suppressed by PRL-1Scer\UAS.cPa.

Expression of PRL-1Scer\UAS.NC partially suppresses the pupal lethality seen when Src64BUY1332 is expressed under the control of Scer\GAL4ap-md544, enabling approximately 70% of the expected adults to eclose.

Expression of BacA\p35GMR.PH in wing discs expressing Src64BUY1332 under the control of Scer\GAL4sd-SG29.1 results in massive tissue overgrowth. Co-expression of bskDN.Scer\UAS suppresses the tissue overgrowth seen in these mutants.

Knockdown of yki, through expression of ykiNIG.4005R under the control of Scer\GAL4sd-SG29.1 prevents Src64BUY1332 induced tissue growth in BacA\p35GMR.PH-expressing wing discs.

While expression of BacA\p35GMR.PH in imaginal wing discs expressing Src64BUY1332 under the control of Scer\GAL4sd-SG29.1 increases disc size up to 2.5 times (compared to controls), co-expression of cpaScer\UAS.N.T:Ivir\HA1 and cpbScer\UAS.cWa in these tissues significantly reduces tissue growth to 1.8 times that found in controls, but does not affect growth of Scer\GAL4sd-SG29.1-->BacA\p35GMR.PH wing discs.
