A genomic fragment from -2702 to +970bp relative to the Ddc transcription start site is fused upstream of the GAL4 coding region. An Hsp70 termination/polyA region and the Ddc exon D 3' untranslated region and 3' flanking sequences (nucleotides +3521 to +4643) are present downstream of the GAL4 coding region. The AUG codon present in the 5' Ddc fragment has been mutated to prevent interference with the translation of GAL4.

Scer\GAL4Ddc.HL9 drives expression in a subset of the dopaminergic PPL2ab neurons and in the majority of the PAM neurons (includes MB-M3 and MB-MP1 neurons).

Scer\GAL4Ddc.HL9 drives expression in 60 neurons of the dopaminergic cluster PAL, 3 neurons of the PAL, 3 neurons of the PPL2ab, 8 neurons of the PPM2 and 1 neuron of the PPM3 cluster. Scer\GAL4Ddc.HL9 labels more neurons of the PAM cluster than Scer\GAL4ple.PF.

Inhibition (using shi1.UAS at 32[o]C) of Scer\GAL4Ddc.HL9 cells in adults does not affect 5 min (single-trial conditioned) aversive taste memory, compared to controls.

Thermogenetic activation (using TrpA1UAS.(B).cKa at 30[o]C) of Scer\GAL4Ddc.HL9 cells in adults, in combination with a paired odor, results in a significant increase in 2-min associative attractive olfactory memory, compared to controls.
