Description

FlyBase curator comment: a 'UAS-myr-Akt1' stock submitted to the Bloomington Drosophila Stock Center by S. Birnbaum was subsequently found to contain an N-terminal deletion in the coding sequence, removing 75% of the PH domain (stock 80935, see FBrf0241238 , personal communication to FlyBase). It is not known whether the 'myrAkt' fly line used in FBrf0238295 corresponds to this deleted construct or whether it contains Akt1 full-length coding sequence.

UAS regulatory sequences drive expression of an active form of Akt1 (protein is tagged with a myristoylation signal). It is not known whether the 'myrAkt' fly line described in FBrf0211161 corresponds to this deleted construct or whether it contains Akt1 full-length coding sequence.

FlyBase curator comment: the UAS-Dakt[myr] fly line used in FBrf0194781 is referenced to FBrf0123227, where a UAS-Dakt[myr] plasmid is transfected into cells. A 'UAS-myr-Akt1' stock submitted to the Bloomington Drosophila Stock Center by S. Birnbaum (an author on FBrf0194781 and FBrf0123227) was subsequently found to contain an N-terminal deletion in the coding sequence, removing 75% of the PH domain (stock 80935, see FBrf0241238 , personal communication to FlyBase). It is not known whether the 'UAS-Dakt[myr]' fly line used in FBrf0194781 corresponds to this deleted construct or whether it contains Akt1 full-length coding sequence.

Posterior dorsocentral bristle neurons expressing Akt1HM04007 under the control of Scer\GAL4pnr-MD237 show a severely increased terminal arbor territory and significantly more synapses (Syt1 puncta) on the contralateral collateral branch, as compared to controls.

The expression of AktUAS.Tag:Myr(Unk) under the control of Scer\GAL4ppl.PP leads to third instar larval salivary glands exhibiting large cells.

Clonal expression of Akt1UAS.Tag:Myr(Unk) under the control of Scer\GAL4Act5C.PP in the larval fat body induces larger cells and larger nuclei, as compared to neighboring control cells.

100% of pupae expressing Akt1Scer\UAS.T:Uuuu\Myr5 under the control of Scer\GAL4hs.PB (using a 30 minute heat shock at 9 hours after puparium formation) still possess intact salivary glands at 20 hours after puparium formation (this is approximately 6 hours after the glands are normally destroyed in wild-type animals).

Larval fat body clones co-expressing Tl10b.UAS.cUa and Akt1UAS.Tag:Myr(Unk) under the control of Scer\GAL4Act5C.PP exhibit the same cell and nucleus size increases as observed upon Akt1UAS.Tag:Myr(Unk) single expression (although the increased nucleus size is partially suppressed), instead of the cell and nucleus size decreases observed upon Tl10b.UAS.cUa single expression.

Co-expression of Akt1Scer\UAS.T:Uuuu\Myr5 rescues the reduced size of larvae and adult escapers expressing Tl10b.Scer\UAS.cUa under the control of Scer\GAL4r4.
