lid10424 mutants are near lethal and the few escapers are morphological normal but short-lived.

lid10424 enhances the variegated phenotype of the w or Sb locus in the In(1)wm4 or T(2;3)SbV background, respectively.

lid10424/lidk06801;lidJmjC.T:Ivir\HA1 'rescue' flies display premature transcriptional reactivation as well as precocious karyosome remodeling (the oocytes display early increase in chromatin openness) in prophase I-arrested oocytes and the eggs show impaired entry into embryogenesis after fertilization resulting in strongly reduced female fertility.

lidk06801/lid10424 mutant female germlines exhibit abnormal morphology of the karyosome, defective clustering of centromeres, and a failure to maintain the synaptonemal complex.

The positional effect variegation (PEV) at the Sb locus caused by T(2;3)SbV is significantly enhanced by single copy of lid10424.

lid10424 homozygotes survive at less than 5% rate, whereas about 50% of lidk06801/lid10424 mutants survive.

lidk06801/lid10424 mutant flies have irregularly shaped mitochondria that are larger than those of wild-type flies and the flies display reduced climbing ability.

lid10424/Df(2L)BSC184 is pupal lethal, with rare adult escapers. There is a significant decrease of germline stem cell (GSC) number, the majority of GSCs appear to be detached from the hub cells, and average hub area is significantly larger in lid10424/Df(2L)BSC184 third instar larval testes, compared to wild type.

There is a significant decrease in mitotic activity of GSCs (decreased proliferation and premature differentiation) in lid10424/Df(2L)BSC184 third instar larval testes compared to wild type. Spermatogonial cysts containing up to four interconnected germ cells in direct association with the hub region are detected in approximately one third of lid10424/Df(2L)BSC184 third instar larval testes and differentiated spermatogonial cysts in direct contact with hub cells are detected. No disintegrating fusome remnants are detected in GSCs from lid10424/Df(2L)BSC184 third instar larval testes and the percentage of GSCs with misoriented centrosomes is not significantly different from wild type.

lid10424/+ enhances the variegation at the w locus which is seen in the In(1)wm4h chromosome and enhances the variegation at the mutant Sb locus which is seen in the T(2;3)SbV chromosome.

Heterozygous lid10424 mutants show normal locomotor activity under a 12 hours of light:12 hours of dark (LD) cycle and total activity is equivalent to that of wild type. Under constant darkness (DD) lid10424/+ flies exhibit disrupted circadian activity rhythms, with the majority of flies showing either no or weak circadian activity rhythms. In flies that do show detectable rhythms, the period is significantly shortened.

Heterozygous flies show strong enhancement of position effect variegation of the w gene caused by In(1)wm4h.

lidk06801/lid10424 animals eclose at approximately half the expected frequency.

Some homozygotes and lidk06801/lid10424 flies appear to die before hatching, although no obvious defects in the larval cuticle are seen. Most homozygotes appear to die at an early pupal stage. Third instar homozygotes often have a small optic brain lobe and small imaginal discs.

Combination with a single copy of lid10424 does not modify the frequency of eye tissue to antenna-like malformed structures transformation in adult flies expressing TAF1BdsRNA.Scer\UAS under the control of Scer\GAL4ey.200.Exel nor the massive eye overgrowth in flies expressing ykiS168A.Scer\UAS.T:Avic\GFP-EGFP,T:Ivir\HA1 with Scer\GAL4GMR.PF or the disrupted wing vein phenotype of flies expressing smo5A.Scer\UAS.T:Zzzz\FLAG driven by Scer\GAL4C-765.

Stat92E06346/+ enhances germline stem cell (GSC) premature differentiation in lid10424/Df(2L)BSC184 third instar larval testes, with spermatocytes found in direct association with hub cells.

Stat92EScer\UAS.cPa driven by Scer\GAL4nos.PU suppresses the GSC loss and premature differentiation (spermatogonial cysts next to the hub) seen in lid10424/Df(2L)BSC184 third instar larval testes.

shgScer\UAS.T:Avic\GFP-rs driven by Scer\GAL4nos.PU suppresses the reduced number of germline stem cells (but not the reduced mitotic activity) seen in lid10424/Df(2L)BSC184 third instar larval testes.

Presence of Ptp61Fc05292/+ suppresses the loss of germline stem cells seen in lid10424/Df(2L)BSC184 third instar larval testes.

lid10424/+ partially suppresses the held-out wing phenotype and largely suppresses the extra wing vein phenotype which is seen in Su(var)3-3ΔN homozygotes.

lid10424/+ partially suppresses the follicle cell defects seen in Su(var)3-3ΔN females.

Su(var)3-91/+ suppresses the ability of lid10424/+ to enhance the variegation at the w locus which is seen in the In(1)wm4 chromosome.

The wing vein truncation phenotype which is seen in NAx-16/+ flies is enhanced by lid10424/+.

The small, rough eye phenotype caused by expression of asf1Scer\UAS.cMa under the control of Scer\GAL4ey.PH is significantly suppressed by lid10424/+.

The dmP0 ; lidk06801/lid10424 combination results in almost complete lethality.

lid10424 ash122 double heterozygotes show T3 to T2 transformations.

Comments

The premature transcriptional reactivation as well as precocious karyosome remodeling (the oocytes display early increase in chromatin openness) in prophase I-arrested lid10424/lidk06801 transheterozygous oocytes is rescued by combination with lidT:Ivir\HA1.

Expression of lid+mDa rescues the locomotor rhythm defects seen in homozygous lid10424 mutant flies.

Expression of lidgJmjC.mut partially rescues the locomotor rhythm defects seen in homozygous lid10424 mutant flies. Under LD (12 hours of light:12 hours of dark) and DD conditions (constant darkness) flies exhibit attenuated day:night differences in activity, with flies showing increased activity around midday and reduced dark anticipation.
