Adulthood-only expression of Atg9HMS01246 under the control of Scer\GAL4esg.PU leads to a significant increase in the number of mitotic cells (phospho-H3 staining) in the adult midgut, as compared to controls. (The temporal regulation of expression is dependent on Gal80[ts] and induced by temperature shift to 29[o]C for 5-6 days during adulthood).

The expression of Atg9HMS01246 under the control of Scer\GAL4Cg.PA blocks the starvation-induced accumulation of acidic vesicles (LysoTracker-positive) in the early third instar larval fat body, as compared to controls. Under standard conditions, there is no change in the number of autophagosome-like structures (Atg8a-positive), as compared to controls.

Constitutive expression of Atg9HMS01246 under the control of Scer\GAL4Myo31DF-NP0001, or adulthood-only expression under the combined control of either Scer\GAL4tub.PU and Gal80[ts] or Scer\GAL4Myo31DF-NP0001 and Gal80[ts], leads to pronounced defects in the midgut, namely a severe increase in cell size and abnormal cell shape, as compared to controls. Adulthood-only expression under the combined control of Scer\GAL4tub.PU and Gal80[ts] leads to intestinal barrier disfunction (as shown by the abnormal permeability to a non-absorbable food dye), leads to the failure to induce autophagy (assessed by the lysosome and autophagosome marker, Lysotracker) in the adult fat body in response to starvation, and leads to a decreased lifespan, as compared to controls.

Constitutive expression under the control of Scer\GAL4Dl-05151-G or Scer\GAL4Su(H).GBE, or adulthood-only expression under the combined control of Scer\GAL4how-24B and Gal80[ts], has no obvious effect in the midgut, as compared to controls.

Clonal expression of Atg9HMS01246 under the control of Scer\GAL4Act.PU has no significant effect on the cell size within larval wing disc or larval eye disc clones, as compared to controls.

The increased midgut cell size resulting from the expression of Atg9HMS01246 under the control of Scer\GAL4Myo31DF-NP0001 is suppressed by the single co-expression of Atg1Scer\UAS.cSa, S6kKQ.Scer\UAS, TorTED.Scer\UAS, InRHMS03166, Pi3K92EHMC05152, Akt1HMS00007 or Patj[Scer\UAS. T:Hsap \MYC], or by the combined co-expression of Tsc1Scer\UAS.cSa and gigScer\UAS.cSa. Conversely, this phenotype is moderately enhanced by the co-expression of Akt1Scer\UAS.Exel. However, this phenotype is not significantly affected by the co-expression of RhebScer\UAS.cPa, Tsc1GD11836, gigGD1454 or Pi3K92ECAAX.Scer\UAS.

The co-expression of Atg9HMS01246 and PatjKK110089 under the control of Scer\GAL4Myo31DF-NP0001 does not enhance the increased midgut cell size resulting from either single expression.
