There is an increase in the frequency of 'winner' clone splitting (used as a readout for winner-loser cell mixing) when cell competition is induced in the wing disc through generation of wild type clones in RpS3Plac92/+ (a Minute mutant) tissues compared with wild type clones in wild type tissue. Clone compactness is also reduced. F-actin levels are higher in wild type clones than in RpS3Plac92/+ cells.

Heterozygous RpS3Plac92 mutant females exhibit a dilated cardiomyopathy phenotype, including an increased end-systolic dimension (EDD) and a decreased fractional shortening (FS) compared to controls. Bristles appear short and thin compared to wild type (Minute phenotype).

Larval development is prolonged by about 51 hours. Female fertility (egg production rate) and vitality (fraction of hatched eggs appearing as adults) is reduced by about 45%. Scutellar bristles of heterozygotes are reduced in length and thickness by about 40%. RpS3Plac92 is lethal in combination with RpS3prv9 and in combination with RpS3prv11 exhibits an extreme/semi-lethal Minute phenotype with a vitality of only 3-5%.

Cell death is similar in the posterior compartment expressing DroncCARD.Scer\UAS (via Scer\GAL4en.PU) to the control anterior compartment in RpS3Plac92/+ wing discs in which wild type clones have been induced. However, some homozygous RpS3Plac92 cell clones are observed.

Scer\GAL4en.PU-mediated expression of both DroncKK104278 and StricaGD12308 together, but neither alone, prevents competitive cell death at RpS3Plac92/+ boundaries with +/+ cells.

Scer\GAL4en.PU-mediated expression of both DroncKK104278 and StricaGD12308 together or DroncKK104278 alone, but not StricaGD12308 alone allows survival of RpS3Plac92 homozygous cell clones.

RpS3Plac92 complements the lethality seen in DNApol-εpl10R.

Scer\GAL4en.PU-mediated expression of BacA\p35Scer\UAS.cHa rescues the normal death of homozygous RpS3Plac92 cell clones in the wing disc. In this background, homozygous RpS3Plac92 clones generated by a 10-min heat shock (inducing 10 recombination events per compartment, on average) has no detectable effect on wing size. By contrast, homozygous RpS3Plac92 clones generated by a 60-min heat shock (resulting in 30 'undead' clones in the posterior compartment, on average) results in a significant increase in the size of posterior compartment, detectable in the adult wings.
