Fat body cells in homozygous Cdk43 mutant larvae show reduced mitochondrial abundance compared to controls.

Cdk43/Cdk4k06503 mutant larval brains have normal gross morphology and glial cell numbers.

Mutant flies show a reduction in body weight compared to wild type-controls.

Homozygotes are more sensitive to X rays than wild type or heterozygous sibling controls.

87% of homozygous males survive to adulthood. Homozygous adult males weigh on average only 81% of the weight of control males. The average wing area of hemizygous males is only 94% that of wild type and the number of cells in the wing is 88% that of wild type. The cells in the wing of hemizygous males are slightly increased compared to wild type.

The progeny of Cdk43 germ-line clone carrying mothers have segmentation defects. Most commonly the cuticles of such embryos have no denticle belt A5 and a gap in denticle belt A4. Segmentation defects are more severe in embryos also zygotically homozygous for Cdk43, including reduction of the A2 and A8 denticle bands, fusion of the A6 and A7 bands, and head defects. These embryos also sometimes display other defects such as poor cuticle development and a dorsal hole. Paternally rescued Cdk43 germ-line clone embryos have defective tracheal systems, generally having several disruptions in the main trunk and several branches. lateral secondary branches is also reduced.

only a few percent of eggs laid by Cdk43 progress normally through the syncytial division cycles, while the other embryos are affected by division errors to a very variable extent. Most embryos display highly variable morphological abnormalities during later embryonic stages.

Homozygous flies are slightly smaller, lighter and have a reduced number of cells in the wing (the only tissue where cell number was examined). Homozygous mutant clones are only about two-thirds of the area, on average, of wild-type sister clones.

Cdk43/Cdk4k06503 completely suppresses the glial neoplasia seen in third instar larvae when btl::EgfrScer\UAS.T:λ\cI-DD and Pi3K92EScer\UAS.T:Hsap\MYC,T:Hsap\CAAX are co-expressed under the control of Scer\GAL4repo.PU.

Overgrowth of HphEP3200; Scer\GAL4Act5C.PP or HphScer\UAS.cFa; Scer\GAL4Act5C.PP somatic clones in third instar wing discs is unaffected by Cdk43/Cdk43.

The wing overgrowth phenotype caused by expression of banEP3622 under the control of Scer\GAL4Bx-MS1096 is not altered by Cdk43/Cdk43. The eye phenotype caused by expression of exScer\UAS.cBa under the control of Scer\GAL4hs.2sev is not altered by Cdk43/+.

When tkvQ253D.Scer\UAS.cNc is driven by Scer\GAL4Act5C.PP in a Cdk43 background, no suppression of the proliferation phenotype is seen.

Expression of both Tsc1Scer\UAS.cTa and gigScer\UAS.cTa in the eye under the control of Scer\GAL4ey.PH results in a much smaller eye than normal. Addition of one copy of Cdk43 enhances the phenotype.

The addition of CycAunspecified, CycBunspecified, CycB3unspecified or cdc2unspecified have no effect on survival of Cdk43 homozygotes.

The addition of Cdk4Scer\UAS.cMa driven by Scer\GAL4da.G32 rescues the weight, body and cell size phenotypes seen in Cdk43 flies. Whilst similar expression of Cdk4D175N.Scer\UAS.T:Hsap\MYC has no effect. Heterozygosity in CycEunspecified is tolerated in Cdk43 heterozygotes, but leads to complete lethality in Cdk43 homozygotes. Cdk43 homozygotes also heterozygous for Rbf11 (and probably Rbf14) are found to be significantly more fertile than Cdk43 homozygotes alone.
