The testis specificity index was calculated from modENCODE tissue expression data by Vedelek et al., 2018 to indicate the degree of testis enrichment compared to other tissues. Scores range from -2.52 (underrepresented) to 5.2 (very high testis bias).

unc-45 protein is detected on western blots at all stages tested. At two hours after egg laying, it localizes to the embryonic blastoderm and co-localizes with non-muscle myosin II. At 14hr AEL, localization is similar to muscle myosin, which is expressed strongly in skeletal muscles such as body-wall muscles and pharyngeal muscle. Weaker staining is observed in gut muscle and in the ectoderm. unc-45 protein was also observed in other skeletal muscles examined including embryonic dorsal tube, adult body-wall muscle, adult dorsal tube, and adult indirect flight muscle.

For each fully sequenced cDNA the DGRC maintains various forms of the cDNA (e.g tagged or untagged) in several different host vectors for subsequent cloning and expression in Drosophila and Drosophila cell lines.
Embryonic injection of dsRNA made from templates generated with primers directed against this gene results in an entirely normal embryonic muscle pattern. However, these muscles never contract (compared to age-matched controls).
