Description

Sequence encoding a GAL4 driver has been inserted at the site of the normal translation initiation codon of the Gr33a locus (a mini-w marker has also been inserted in addition to the GAL4 coding region and a portion of the endogenous Gr33a coding region has been replaced by the inserted sequences).

ScerGAL4Gr33a-GAL4 drives expression in bitter-sensing neurons in the larval dorsal pharyngeal organ.

Drives expression in bitter-activated gustatory receptor neurons in the adult labellum and in the tarsal segments of legs.

ScerGAL4Gr33a-GAL4 drives expression in S-a and S-b bitter-activated gustatory receptor neurons in the adult labellum.

ScerGAL4Gr33a-GAL4 drives expression in L-canavanine-insensitive S2- and I-type sensilla.

ScerGAL4Gr33a-GAL4 drives expression in all I-type and S-type sensilla, which represent a total of 22 gustatory receptor neurons in each labellum.

ScerGAL4Gr33a-GAL4 drives expression in two of the three gustatory receptor neurons innervating the dorsolateral terminal organ. These neurons coexpress ppk23, but not ScerGAL4GMR58F10

ScerGAL4Gr33a-GAL4 drives expression in about 20 gustatory receptor neurons (GRNs) in the adult labellum; these GRNs coexpress Gr66a. ScerGAL4Gr33a-GAL4 also drives expression in multiple gustory receptor neurons in the tarsal segments of the legs; these include 7 in each prothoracic leg, 4 in each mesothoracic leg, and 4 in each methoracic leg. Several of the prothoracic GRNs coexpress ScerGAL4Gr32a.3.776.

Detailed Description

Inhibition (using Ctet\tetXTNT-LC.UAS or Hsap\KCNJ2UAS.EGFP) of Scer\GAL4Gr33a-GAL4 cells in adults results in a significant increase in proboscis extensions in response to higher concentrations of hexanoic acid, with no effect on the response to sucrose, compared to controls.

Inhibition (using Hsap\KCNJ2UAS.EGFP) of the combined set of Scer\GAL4Gr33a-GAL4 and Scer\GAL4Gr64f.9.7 cells in adult females does not affect preference for oviposition on acid-containing medium, compared to controls.

Males in which neurons expressing Gr33aGAL4 have been silenced show no difference in courtship intensity as compared with wild type or controls, but lose preference for younger females, instead showing no significant preference for either younger or older females.

Inhibition (using Ctet\TeTxLCTNT.UAS) of Scer\GAL4Gr33a-GAL4 cells does not affect the preference of larvae for larval-treated substrate, compared to controls.
