png2/png5 females produce two classes of embryos 30-55% are "multinucleate" (indicating that some mitotic division has occurred), containing more than 5 giant polyploid nuclei. The remaining embryos contain up to 5 giant polyploid nuclei (indicating a lack of mitotic divisions). Homozygous females produce 5-9% of "multinucleate" embryos.

The meiotic products and pronuclei of embryos derived from homozygous mothers overreplicate.

Homozygous females lay eggs in which the meiotic products and the pronucleus undergo extensive DNA replication resulting in a 'giant nuclei' phenotype. dhdJ5 does not suppress the premature initiation of DNA synthesis in double mutants.

Unfertilised eggs from a homozygous mother initiate DNA replication inappropriately and in fertilised eggs DNA replication is uncoupled from nuclear division, both unfertilised and fertilised eggs have one giant polyploid nuclei. Chromosomes in the embryos are never condensed and do not appear polytene. Premature replication begins at the earliest during passage into the uterus. Trans-heterozygotes with the multiple nuclei alleles, png2 and png4, produce embryos with multiple polyploid nuclei.

Embryonic defects detectable during preblastoderm stages.
