Bj1 transcripts are abundant in early embryos and adult females and are barely detectable at other developmental stages. Bj1 transcripts are strongly enriched in the prospective oocyte of young follicles and only low levels are present in nurse cells. At stage 10 of oogenesis, strong expression starts in the nurse cells and the transcripts are later transported to the oocyte. Bj1 transcripts are uniformly distributed in early embryos. In later embryos, transcripts persist only in neuroblasts and ganglion cells derived from them.

Bj1 protein is found exclusively in nuclei in the oocyte and is predominantly localized to nuclei in early cleavage stage embryos. From the blastoderm stage onwards, Bj1 protein is exclusively nuclear during interphase. During mitosis, it becomes evenly distributed throughout the cells. It is found in all bands of polytene chromosomes and is absent from developmental puffs, heat-shock puffs and interbands.
Bj1 is maternally supplied to early embryos; the maternally-supplied protein can be detected in the nuclei of late-stage Bj1-null embryos. In wild-type embryos, Bj1 protein expression remains high in the CNS after embryonic stage 13, but is sharply decreased in other tissues. Bj1 is found exclusively in the nucleus during interphase, but colocalizes with mitotic chromosomes and forms spindle-like structures during mitosis.
Bj1 (RCC1) protein wasassociated with chromatin throughout the cell cycle including during mitosis in methanol fixed embryos and as monitored by injected rhodamine labeled human RCC1 protein.

Alleles of Bj1 show general defects in lateral CNS development.
Mutants show supernumerary pigment cells between ommatidia, evident in the adult as a rough eye due to a disruption of the normally precise ommatidial pattern. This phenotype results from a failure of programmed cell death in the retinal epithelium.
The maternally expressed Bj1 gene product binds to chromatin to establish or maintain a proper higher order structure as a prerequisite for regulated gene expression. Disruption of the structure may cause both misexpression and default repression of genes.
