Gene Summary
screw (scw) encodes a BMP-type ligand that forms a heterodimer with the product of dpp. The heterodimer is required to achieve the peak levels of BMP signal in the dorsal-most cells of the early blastoderm embryo that are necessary to specify the amnioserosa. Gene Snapshot
screw (scw) encodes a BMP-type ligand that forms a heterodimer with the product of dpp. The heterodimer is required to achieve the peak levels of BMP signal in the dorsal-most cells of the early blastoderm embryo that are necessary to specify the amnioserosa. [Date last reviewed: 2019-03-14]
Gene Group (FlyBase)
BONE MORPHOGENETIC PROTEINS -
Bone Morphogenetic Proteins (BMPs) are a subfamily of the Transforming Growth Factor (TGF)-β family - a group of extracellular ligands for the TGF-β-Receptor family. The binding of a BMP dimer to a heterodimeric serine/threonine kinase receptor complex results in the phosphorylation of Mad, a member of the Smad family. Mad forms a complex with the co-Smad, Med. This complex translocates into the nucleus and regulates the transcription of target genes in concert with other nuclear cofactors. (Adapted from FBrf0236482.)
Pathway (FlyBase)
BMP SIGNALING PATHWAY CORE COMPONENTS -
The Bone Morphogenetic Protein (BMP) signaling pathway is one of two branches of Transforming Growth Factor-β family signaling in Drosophila. The binding of a BMP family dimer to a heterodimeric serine/threonine kinase receptor complex results in the phosphorylation of Mad, a member of the Smad family. Mad forms a complex with the co-Smad, Med. This complex translocates into the nucleus and regulates the transcription of target genes in concert with other nuclear cofactors. (Adapted from FBrf0236482.)
Protein Function (UniProtKB)
Part of the signal that specifies dorsal cell fates in the embryo. Acts together with dpp.
(UniProt, P54631)
Phenotypic Description (Red Book; Lindsley and Zimm 1992)
scw: screw
Embyonic lethal; partially ventralized; cephalic furrow shifted dorsally with defects in germ band extension. Ventral denticles extended laterally; spiracles and two to three terminal segments retracted into embryo (Arora and Nusslein-Volhard).
Summary (Interactive Fly)

TGF-ß superfamily - the Scw/dpp heterodimer is required to achieve the peak levels of BMP signal in the dorsal-most cells of the early blastoderm embryo that are necessary to specify the amnioserosa.

If you don't see a structure in the viewer, refresh your browser.
Model Confidence:
  • Very high (pLDDT > 90)
  • Confident (90 > pLDDT > 70)
  • Low (70 > pLDDT > 50)
  • Very low (pLDDT < 50)

AlphaFold produces a per-residue confidence score (pLDDT) between 0 and 100. Some regions with low pLDDT may be unstructured in isolation.

The scw product negatively regulates dac expression in the embryonic head.

New annotation (CG31695) in release 3 of the genome annotation.

dpp/tkv signaling is obligatorily required, while scw/sax signaling is necessary but not sufficient for dorsal patterning in the embryo.

scw is located between exons 4 and 5 of Lar.

          scw positively modulates dpp activity.

Genetic and phenotypic data reveals that scw is involved in post-transcriptional regulation of dpp activity by elevating dpp activity dorsally. Mutations in scw disrupt amnioserosa formation. Injection of dpp RNA into the ventral side does not result in an asymmetric response, a second amnioserosa forms at the site of injection. This suggests that the scw protein is critical to the local response to dpp RNA.

scw and dpp act together to establish distinct response boundaries within the dorsal half of the embryo, perhaps by forming heterodimers that have a higher activity than homodimers of either molecule alone.

scw alleles display relative phenotypic strengths, this may be correlated to the progressive loss of dorsal pattern elements in the ventralised mutants.

          scw has been molecularly characterised.

scw is required for the normal ontogeny of the zen pattern and fating of the amnioserosa.

Involved in the regulatory hierarchy responsible for the asymmetric distribution and function of zygotic regulatory gene products along the DV axis of early embryos. scw mutants exhibit a weak ventralising phenotype in which the maintenance of the zen pattern is affected.

NCBI Gene - Gene integrates information from a wide range of species. A record may include nomenclature, Reference Sequences (RefSeqs), maps, pathways, variations, phenotypes, and links to genome-, phenotype-, and locus-specific resources worldwide.
GenBank Nucleotide - A collection of sequences from several sources, including GenBank, RefSeq, TPA, and PDB.
GenBank Protein - A collection of sequences from several sources, including translations from annotated coding regions in GenBank, RefSeq and TPA, as well as records from SwissProt, PIR, PRF, and PDB.
RefSeq - A comprehensive, integrated, non-redundant, well-annotated set of reference sequences including genomic, transcript, and protein.
UniProt/GCRP - The gene-centric reference proteome (GCRP) provides a 1:1 mapping between genes and UniProt accessions in which a single 'canonical' isoform represents the product(s) of each protein-coding gene.
UniProt/Swiss-Prot - Manually annotated and reviewed records of protein sequence and functional information
UniProt/TrEMBL - Automatically annotated and unreviewed records of protein sequence and functional information
