In in vitro assays, purified native or recombinant Nlp protein coimmunoprecipitates with core histones. In a reaction containing DNA, purified core histones, ATP and partially purified CAF1 fraction, the longer Nlp protein was able to promote assembly of approximately regularly spaced nucleosome arrays, while the shorter Nlp protein could not. When Nap1 and Nlp proteins were compared, only Nap1 was able to promote sperm decondensation in Xenopus laevis sperm chromatin (contrast this with the
results of FBrf0093082).
The shorter Nlp protein lacks 31 N-terminal residues found in the longer protein; this deletion abolishes the specificity of interaction with core histones.
