Overview

Assessed in larval eye discs, loss-of-function mutations of Dmel\Rab5 in clones of cells overexpressing a growth regulator microRNA, mir-ban (bantam), result in dramatic tumor growth and malignant invasion of adjacent tissues, specifically the ventral nerve cord. Neither overexpression of mir-ban alone nor Rab5 mutation alone causes tumor growth or metastatic behavior. This system has been used to assess transcriptome changes over the course of tumor growth and progression.

There are three human genes orthologous to Dmel\Rab5, RAB5A, RAB5B, and RAB5C. UAS constructs of tagged Hsap\RAB5C and Hsap\RAB5A human genes have been introduced into flies, but have not been characterized in the context of this disease model.
