Germline stem cell number (GSC) is significantly reduced in chic11/chic01320 mutant larval testes compared to controls. The number of GSCs is further reduced in newly eclosed adults. Somatic cells move to fill the area adjacent to the hub in the absence of GSCS.

Germline stem cell number is significantly reduced in chic221/chic01320 larval testes compared to controls. In contrast to wild type testes, elongated spermatid bundles are located close to the apical tip in newly eclosed flies. Spermatogonia and spermatocytes are rare or not detected.

Mutant larvae show defects in wound closure.

Developmental changes in stress fibre orientation in chic01320 follicle cells occur as in wild type, though some defective actin organization is observed with the novel occurrence of fan-shaped stress fibres orientated perpendicular rather than parallel to the oocyte A-P axis.

At a stage when the first cells have just invaginated from the salivary gland placode, cell shapes within mutant placodes often appear irregular compared to wild type. At later stages, the salivary glands invaginate but are irregular in shape, and the placodal and surrounding epidermal cells on the surface of the embryos appear disrupted.

Stage 9 chic01320/chic221 oocytes show premature cytoplasmic streaming.

40.5% of eggs laid by heterozygous females are short and round and have smaller and broader dorsal appendages than normal.

The karyosomes of chic01320 mutants are abnormal in around one third of cases.

8% of chic01320/chick13321 egg chambers have border follicle cell migration defects. 16% of chic01320/chic221 egg chambers have border follicle cell migration defects.

Homozygous egg chambers contain binucleate cells. Remnants of ring canals are usually seen, although rarely no ring canal remnants are detectable in binucleate cells.

chicsand-1/chic01320 females fail to lay eggs.

DNA fragmentation occurs in homozygous nurse cells, but is delayed with respect to wild-type.

Homozygous adults have abnormal bristles. Female fertile in transheterozygous combination with chic8A. Onion-stage spermatids of chic01320/chicR1 males usually contain a single Nebenkern of larger than normal size, associated with two or more normal-sized nuclei (64.3% of spermatids have 4 nuclei). Onion-stage spermatids of chic01320/chic8A males usually contain a single Nebenkern of larger than normal size, associated with two or more normal-sized nuclei (65.0% of spermatids have 4 nuclei). 69.5% of asters show irregular positioning and delayed migration during meiotic divisions in chic01320/chicR1 males.

Microtubules are bundled to the cortex of stage 8 egg chambers. Premature cytoplasmic streaming occurs in stage 8 egg chambers. Eggshells exhibit dorsal/ventral patterning defects and often show fused dorsal appendages.

Homozygotes show extra macrochaetae. The egg chambers of chic01320/chic221 and chic01320/Df(2L)GpdhA females usually have too few nurse cells that are mostly binucleate. The variation in nurse cell number is great, with some chambers having as many as 30. 10% of egg chambers show a failure of border cell migration.

Fully penetrant egg chamber phenotype: nurse cells fail to deliver all their cytoplasm to the oocyte resulting in small oocytes surrounded by an egg shell. Strong allele of chic.

Expression of shgScer\UAS.T:Avic\GFP-rs in germ cells under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 does not suppress the germline stem cell (GSC) loss seen in chic11/chic01320 mutant testes.

Expression of shgScer\UAS.T:Avic\GFP-rs in germ cells under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 does not suppress the germline stem cell (GSC) loss seen in chic221/chic01320 mutant testes.

Expression of Apc2Scer\UAS.T:Avic\GFP in germ cells under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 suppresses the germline stem cell (GSC) loss seen in chic11/chic01320 mutant testes.

Expression of Apc2Scer\UAS.T:Avic\GFP in germ cells under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 does not suppress the germline stem cell (GSC) loss seen in chic221/chic01320 mutant testes.

A chic01320 heterozygous background partially suppresses the rough eye phenotype seen in Fmr1sev.PW mutants.

Expression of capuΔN.Scer\UAS.P\T.T:Avic\GFP under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 does not suppress the premature cytoplasmic streaming seen in stage 9 chic01320/chic221 oocytes.

Only 13.4% of eggs laid by chic01320/+ spoon1/+ double heterozygous females have a mutant phenotype.

80% of chic01320/chick13321 ; th5/+ egg chambers have border follicle cell migration defects. 51% of chic01320/chick13321 ; th21-2s/+ egg chambers have border follicle cell migration defects. In some egg chambers, the border follicle cells migrate to the side of the egg chamber instead of through the middle of the nurse cells, a phenotype that is never seen in wild-type egg chambers. 77% of chic01320/chic221 ; th5/+ egg chambers have border follicle cell migration defects. 48% of chic01320/chic221 ; th21-2s/+ egg chambers have border follicle cell migration defects.

S48-5/chic01320 mutants show 21.90%+-5.89 misrotated ommatidia compared to 9.55%+-1.43 seen in S48-5 mutants alone.

The phenotype caused by expression of msnEP549 under the control of Scer\GAL4GMR.PF is not modified by the addition of chic01320.

6% of the eggs from capu2/capu2F and 30% from chic01320/chic01320 have fused dorsal appendages, whereas 76% of those from capu2, chic01320/capu2F, chic01320 have dorsal/ventral defects. Similarly capu2F, chic01320/capu2F, chic01320 produce 70% eggs with dorsal/ventral defects while capu2F/capu2F alone produced only 2%.
