    dallygem mutant flies exhibit a reduction in the number of chemosensory bristles. In dallygem heterozygous mutants, the average number of chemosensory bristles is slightly lower than wild-type, but the difference is not statistically significant.

    dallygem mutant animals display a much reduced number of chemosensory and mechanosensory bristles at the anterior dorsal wing margin compared to wild-type. No ectopic mechanosensory bristles are observed.

    The ovarioles of homozygous females show progressive loss of the germline stem cells (GSCs), with 91% of ovarioles containing no GSCs in the niche at day 10 after eclosion. The number of cap cells in the niche is normal in the mutant ovarioles. Programmed cell death is not detected in cap cells or germline cells in the mutant germaria.

    dallygem flies show a reduction in the number of chemosensory bristles at the anterior margin of the wing compared to wild-type flies. The L5 vein in the wings of these flies does not form properly and stops short between the posterior crossvein and the wing margin. These mutants also show a loss of the anterior dorsocentral bristles and the posterior supraalar bristles on the notum. This phenotype is also seen in dallygem/dallyΔP-527 and dallygem/Df(3L)Scf-R6 transheterozygotes.

    Expression of dallyScer\UAS.cJa under the control of Scer\GAL4A9 in a dallygem/dallyΔP-527 or dallygem mutant background results in the formation of excess dorsocentral bristles (the excess is around 30% compared to wild-type wings).

    Wing vein V does not reach the posterior edge of the wing in dallygem mutants.

    In dallygem/dallyΔP-527 adults, the posterior postalar bristle (pPA) is absent from 5% of all individuals. These adults also display defects in wing patterning. In addition to notches in the margin, the wings of these mutants show a truncation of longitudinal vein L5.

    The number of the multiply innervated (recurved) bristles in the dorsal row of dally06464/dallygem wings decreases to 55% of that of wild-type flies. dallyΔP-527/dallygem animals show a loss of both twin campaniform sensilla of the margin (TSM1 and 2) in 70% of animals. In dallygem/dallyΔP-527 flies, 98% of animals lack the posterior supraalar bristle (pSA), 93% the anterior dorsocentral bristles (aDC) and 3% the posterior supraalar bristle (pSA). In dallygem/dally06464 flies, 86% of animals lack the posterior supraalar bristle (pSA), 24% the anterior dorsocentral bristles (aDC) and 1% the posterior supraalar bristle (pSA). In dallygem/dallyΔP-188 flies, 96% of animals lack the posterior supraalar bristle (pSA), 60% the anterior dorsocentral bristles (aDC) and 1% the posterior supraalar bristle (pSA). In the wing disc of dallyΔP-527/dallygem animals, the number of sensory organ mother cells (SMCs) for chemosensory bristles at the anterior wing margin is severely reduced, as compared to wild-type discs. An absence of the SMCs for the aDC, pSA are also observed in these wing discs. The size of the TSM proneural cluster is severely decreased in these mutants, whilst the L3 proneural cluster remains unaffected.

    16% of dallygem/dallyΔP-188 mutant embryos have cuticle defects. About 90% of dallyΔP-188/dallygem flies show abnormalities in their genitalia. About 70% of dally06464/dallygem flies show abnormalities in their genitalia.

    Homozygous males and females display a genitalia defective phenotype, all or parts of the genital disc derivatives are missing. In addition flies have abnormal wings, L5 is incomplete.

    A heterozygous dallygem mutant in a Hsepid12 homozygous background results in a decreased number of chemosensory bristles.

    Hsepid12; dallygem double mutants are lethal.

    The addition of wgl-17 to dallygem/dally06464 flies, leads to enhancement of the posterior supraalar bristle (pSA) phenotype seen in dallygem/dally06464 flies alone: 30% of flies have the phenotype, rather than 1%. The addition of put10460 to dallygem/dally06464 flies, leads to enhancement of the anterior dorsocentral bristle (aDC) phenotype seen in dallygem/dally06464 flies alone: 90% of flies have the phenotype, rather than 24%. The pSA bristle phenotype is also enhanced by the addition of put10460. All flies have the phenotype, rather than 86%. The addition of dallygem/+ enhances the pSA phenotype of Tp(3;3)iro-1 or Df(3L)iro-DFM3/+ flies.

    Heterozygosity for wgl-12 increases the penetrance of cuticle defects in dallyΔP-188/dallygem embryos from 16% to 41%. Embryos heterozygous for dallygem derived from mothers heterozygous for sglN71 show cuticle defects characteristic of a reduction in wg signalling. The abnormal genitalia phenotype of dallyΔP-188/dallygem flies is dominantly suppressed (down to less than 5% penetrance) by wgl-12. The abnormal genitalia phenotype of dally06464/dallygem flies is dominantly suppressed (down to less than 10% penetrance) by wgl-17.

    Expression of dallyΔGAG.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4A9 restores the L5 wing vein in 35% of dallygem homozygotes and 76% of dallygem/dallyΔP-527 transheterozygotes. It performs a slight rescue of the number of chemosensory bristles in dallygem/dallyΔP-527 flies but does not restore posterior supraalar bristles.

    Expression of dallyΔGAG.Scer\UAS under the control of Scer\GAL4A9 partially rescues the phenotypes of dallygem, dallygem/Df(3L)Scf-R6, and dallygem/dallyΔP-527 mutants. The transgene substantially restores chemosensory bristle numbers in the wing margin, almost fully rescues the formation of wing vein L5, almost fully restores the formation of the anterior dorsocentral bristles on the notum without inducing the formation of extra ones for all mutants. However, it fails to restore formation of the posterior supraalar bristles.
