In the larval salivary gland, Hel25E protein associates with the polytene chromosomes. In ovaries, staining is seen in nurse cell and follicle cell nuclei. Confocal microscopy of cycle 14 embyros double-labeled with antibodies against Hel25E and betaTub60D shows that Hel25E disappears from within mitotic domains, although it is present in the nuclei of the other cells at the surface of the embryo.

For each fully sequenced cDNA the DGRC maintains various forms of the cDNA (e.g tagged or untagged) in several different host vectors for subsequent cloning and expression in Drosophila and Drosophila cell lines.
RNAi screen using dsRNA made from templates generated with primers directed against this gene results in chromosome misalignment on the metaphase spindle when assayed in S2 cells. This phenotype can be observed when the screen is performed with or without Cdc27 dsRNA.
Hel25E encodes a putative RNA helicase that contains an unusual variant (DECD) of the DEAD motif common to these proteins.
