Chie5.5/+ mutants exhibit multiple wing notches.

Embryos without maternal and zygotic Chi from mothers with Chie5.5 mutant germline clones do not develop, the few rare escapers show a lawn of ventral denticles.

Heterozygotes display duplication of scutellar bristles with <20% penetrance.

Clones homozygous for Chie5.5 form ectopic eye tissue in the ventral head capsule.

Heterozygous flies have extra dorsocentral bristles.

Projection neurons that are part of Chie5.5 homozygous somatic clones fail to target the correct glomeruli and end up occupying the inappropriate glomeruli. In the case of clones affecting antero-dorsal projection neurons, a fraction of the affected dendrites are mis-targeted to the subesophageal ganglion.

Flies carry somatic clones homozygous for Chie5.5 in their legs have defects in all segments including fusion of tarsal segments 4 & 5 and loss of the unguis (claw).

Heterozygotes are morphologically normal.

Clones in the wing disc induced in the second larval instar stage can cause a bifurcation and outgrowth of the dorsal wing pouch. Clones are comparable in size to their twin spots in both the dorsal and ventral compartments. Dorsal clones in the wing induced in the second larval instar stage induce ectopic wing margin structures along their borders and induce outgrowth of the dorsal compartment of the wing. Ventral clones produce no phenotype. Dorsal clones in the wing induced in early third larval instar stage (72 hours after egg laying) at or near the endogenous margin induce ectopic wing margin tissue. Clones at a distance from the wing margin induce blistering, without inducing ectopic wing margin. Mutant cells in clones in the dorsal compartment at the anterior wing margin differentiate ventral margin identity. Clones in the wing induced in mid third larval instar stage (96 hours after egg laying) do not induce ectopic margin.

Chie5.5 homozygotes show pathfinding errors in the neurons that normally express ap; the neurons are highly defasciculated, choosing inappropriate pathways within the connectives. The projections of tup-expressing intersegmental nerve motor neurons show no clear alterations compared to wild type in homozygous embryos.

Heterozygotes have normal wings. Embryos derived from germ-line clones fail to hatch, cuticles exhibit normal dorsal-ventral polarity but severe segmentation defects. Phenotype is less severe at 25oC than 18oC. Wild type paternal Chi does not substantially alter development.

Homozygotes develop more slowly, are smaller and less active that heterozygous siblings. Chie5.5/Chig1 transheterozygotes die as larvae.
