Expression of Hsap\FOXO1::Hsap\PAX7Scer\UAS.cGa under the control of Scer\GAL4Mhc.PW is semi-lethal at 23[0]C.

Flies harbouring one copy of both Hsap\FOXO1A::Hsap\PAX7Scer\UAS.cGa and Scer\GAL4Mhc.PW exhibit either larval or pupal lethality, depending on the particular Hsap\FOXO1A::Hsap\PAX7Scer\UAS.cGa insertion line used.

Larvae expressing Hsap\FOXO1A::Hsap\PAX7Scer\UAS.cGa under the control of Scer\GAL4Mhc.PW exhibit morphologically normal musculature up to day 2 of larval life. However, early third instar larvae exhibit abnormal muscle morphology: many individual myofibers are absent, with all larval muscle groups appearing susceptible (although in a random distribution from animal to animal), and surviving myofibers appear wispy and hypotrophic.

Expression of Hsap\FOXO1A::Hsap\PAX7Scer\UAS.cGa under the control of Scer\GAL4Mhc.PW generates nucleated myogenic-type cells from differentiated, syncitial myofibers in larvae. Hsap\FOXO1A::Hsap\PAX7Scer\UAS.cGa-expressing cells then disseminate and infiltrate non-native tissue compartments, including the larval CNS.

Co-expression of Hsap\FOXO1A::Hsap\PAX7Scer\UAS.cGa and Ras85DV12.Scer\UAS under the control of Scer\GAL4Mhc.PW results in disorganized larval musculature: the contours of many myofibres are vague and ill-defined. Collections of mononuclear cells are present (at the expense of syncitial myofiber tissue) at a significantly higher frequency than in larvae expressing Hsap\FOXO1A::Hsap\PAX7Scer\UAS.cGa alone.

Heterozygosity for Ras85De2F, Ras85De1B or Ras85D06677 suppresses the larval muscle phenotype seen in animals expressing Hsap\FOXO1A::Hsap\PAX7Scer\UAS.cGa under the control of Scer\GAL4Mhc.PW and also results in viable adult escapers. Of these three alleles, Ras85De2F is the strongest suppressor: at the lower temperature of 23 oC, 83% of Hsap\FOXO1A::Hsap\PAX7Scer\UAS.cGa, Scer\GAL4Mhc.PW, Ras85De2F/+ larvae are rescued to adult viability whereas only 15% of Hsap\FOXO1A::Hsap\PAX7Scer\UAS.cGa, Scer\GAL4Mhc.PW larvae survive to adulthood.
