New stable cell line derived from OSC : An OSC cell line stably expressing flag-tagged Spyo\Cas9 was generated. Additionally, new stable cell lines were derived from the OSC[Cas9] cell line: OSC cell lines stably expressing HA-tagged Dora, and a knockout of Dora in which a hygromycin resistance gene was inserted into its open reading frame, were generated. New stable cell lines were then generated from the Dora and Dora-KO: cell line stably expressing Rren\LUC fused to the 3'UTR of Tom.

The gene is named 'Dorado' after a fish that preys on octopuses from the Argonauta genus, as the gene and its orthologs are proposed to mediate target-directed miRNA degradation (TDMD) through the ubiquitination of an Argonaute protein.
