Gene Summary
Core component of the splicing-dependent multiprotein exon junction complex (EJC) deposited at splice junctions on mRNAs (PubMed:14973490, PubMed:24967911). Involved in exon definition of genes containing long introns, including the rolled/MAPK gene (PubMed:20946982, PubMed:20946983). The mago-tsu heterodimer interacts with the EJC key regulator Pym leading to EJC disassembly in the cytoplasm (PubMed:24967911). Has a role in oskar mRNA localization to the posterior pole of the developing oocyte (PubMed:11691839). (UniProt, Q9V535)

Protein Family (UniProt)
Belongs to the RBM8A family. (Q9V535)
Summaries
Gene Group (FlyBase)
SPLICEOSOME COMPLEX C -
Nuclear pre-mRNA splicing is catalyzed by the spliceosome which assembles in a step-wise manner. Complex B undergoes a number of structural rearrangements and U4 and U1 snRNPs dissociate to generate complex C, the catalytic spliceosome. (Adapted from PMID:24452469 and PMID:23118483).
SPLICEOSOME COMPLEX P -
Nuclear pre-mRNA splicing is catalyzed by the spliceosome which assembles in a step-wise manner. Complex P, the post-spliceosomal complex, is formed after the catalytic removal of the intron by complex C. Complex P contains the lariat intron and spliced exons. (Adapted from PMID:24452469 and PMID:23118483).
Protein Function (UniProtKB)

Structure
Protein 3D structure   (Predicted by AlphaFold)   (AlphaFold entry Q9V535)

If you don't see a structure in the viewer, refresh your browser.
Model Confidence:
  • Very high (pLDDT > 90)
  • Confident (90 > pLDDT > 70)
  • Low (70 > pLDDT > 50)
  • Very low (pLDDT < 50)

AlphaFold produces a per-residue confidence score (pLDDT) between 0 and 100. Some regions with low pLDDT may be unstructured in isolation.

Subunit Structure (UniProtKB)

Heterodimer (via RRM domain) with mago (PubMed:11691839, PubMed:12704080, PubMed:12730685). Part of the mRNA splicing-dependent exon junction complex (EJC) complex; the core complex contains btz/CASC3, eIF4AIII, mago and tsu/RBM8A (PubMed:14973490). Interacts with Pym (via N-terminus); the interaction is direct (PubMed:14968132, PubMed:24967911). Interacts with eIF4AIII (PubMed:14973490).

(UniProt, Q9V535)
Domain

The RNA recognition motif (RRM) is involved in the interaction with mago. The RNA-binding activity of such domains is therefore uncertain.

Testis-specificity index

The testis specificity index was calculated from modENCODE tissue expression data by Vedelek et al., 2018 to indicate the degree of testis enrichment compared to other tissues. Scores range from -2.52 (underrepresented) to 5.2 (very high testis bias).

Please Note This section lists cDNAs and ESTs that fall within the genomic extent of the gene model, which may include cDNAs and ESTs of genes within introns, or of overlapping genes. Please see JBrowse for alignment of the cDNAs and ESTs to the gene model.

For each fully sequenced cDNA the DGRC maintains various forms of the cDNA (e.g tagged or untagged) in several different host vectors for subsequent cloning and expression in Drosophila and Drosophila cell lines.

tsu is required to restrict the oocyte fate to a single cell during oogenesis.

The mago/tsu physical interaction is conserved, occurs in vitro and in vivo and is not RNA mediated. mago and tsu gene products localize within nuclear speckles. mago/tsu heterodimers associate with spliced mRNAs carrying an exon-exon junction complex.

mago and tsu are required for cell viability.
