Gene Summary
Wnk kinase (Wnk) encodes a chloride-sensitive Ser/Thr kinase that signals through a downstream target Ser/Thr kinase, encoded by fray, which is activated by phosphorylation the product of Wnk. Its roles include regulation of ion transport and cell growth, and differentiation and developmental patterning, including roles in canonical Wnt signaling and induction of the product of Awh. Gene Group (FlyBase)
UNCLASSIFIED CONVENTIONAL PROTEIN KINASE DOMAIN KINASES -
This group comprises conventional protein kinase domain kinases that do not classify under other groups in FlyBase.
Pathway (FlyBase)
POSITIVE REGULATORS OF WNT-TCF SIGNALING PATHWAY -
Positive regulators of Wnt-TCF (canonical Wnt) signaling up-regulate the pathway, resulting in the enhancement of transcriptional regulation mediated by β-catenin (arm).
Protein Function (UniProtKB)
Serine/threonine-protein kinase component of the WNK-SPAK/OSR1 kinase cascade, which plays an important role in the regulation of electrolyte homeostasis and regulatory volume increase in response to hyperosmotic stress (PubMed:23797875, PubMed:25086033, PubMed:36318922). Wnk mediates regulatory volume increase in response to hyperosmotic stress by acting as a molecular crowding sensor, which senses cell shrinkage and mediates formation of a membraneless compartment by undergoing liquid-liquid phase separation (PubMed:36318922). The membraneless compartment concentrates Wnk with its substrate Fray, promoting Wnk-dependent phosphorylation and activation of downstream kinase Fray. Following activation, Fray catalyzes phosphorylation of ion cotransporters Ncc69 and Irk1, regulating their activity (PubMed:25086033, PubMed:35303418, PubMed:36318922). Phosphorylation of Na-K-Cl cotransporter Ncc69 promotes its activation and ion influx (PubMed:25086033). Involved in circadian rhythms in small ventral lateral (sLNv) pacemaker neurons: in the morning, Wnk activity is repressed by high levels of intracellular chloride; in contrast Wnk activation in the evening promotes the activation of the inwardly rectifying potassium channel Irk1 via Fray (PubMed:35303418). Acts as a positive regulator of the canonical Wnt signaling pathway during wing disk development (PubMed:23797875).

Structure
Protein 3D structure   (Predicted by AlphaFold)   (AlphaFold entry M9PGC5)

If you don't see a structure in the viewer, refresh your browser.
Model Confidence:
  • Very high (pLDDT > 90)
  • Confident (90 > pLDDT > 70)
  • Low (70 > pLDDT > 50)
  • Very low (pLDDT < 50)

AlphaFold produces a per-residue confidence score (pLDDT) between 0 and 100. Some regions with low pLDDT may be unstructured in isolation.

Please Note This section lists cDNAs and ESTs that fall within the genomic extent of the gene model, which may include cDNAs and ESTs of genes within introns, or of overlapping genes. Please see JBrowse for alignment of the cDNAs and ESTs to the gene model.

cDNA clones, fully sequenced
BDGP DGC clones
Other clones
Drosophila Genomics Resource Center cDNA clones

For each fully sequenced cDNA the DGRC maintains various forms of the cDNA (e.g tagged or untagged) in several different host vectors for subsequent cloning and expression in Drosophila and Drosophila cell lines.

RNAi generated by PCR using primers directed to this gene causes a cell growth and viability phenotype when assayed in Kc167 and S2R+ cells.

dsRNA made from templates generated with primers directed against this gene tested in RNAi screen for effects on Kc167 and S2R+ cell morphology.
