The Pi3K21B adaptor protein was purified from
S2 cells as a heterodimeric complex with Pi3K92E protein. The Pi3K21B
protein sequence includes two SH2 domains but lacks the SH3 and BH domains
of mammalian adaptor counterparts. As in the mammalian adaptors, the
inter-SH2 domain is predicted to have a coiled-coil structure. The
Pi3K92E/Pi3K21B protein complex possesses lipid and protein kinase
activity when affinity purified from S2 cells.

Df(2L)Pi3K21B-A/Df(2L)Pi3K21B-B, P{U2af38.gR10} mutants (null for Pi3K21B) stop growing normally early in the third larval instar. Though they continue to move and feed for approximately 20 days they grow very slowly. Larvae are transparent, have small imaginal discs and have reduced fat body. Nervous system, trachea and gut appear normal. Some form pupal cases but show no indication of pupal development. Mutant clones in the eye reveal that Pi3K21B is autonomously required for adult cells to reach their normal size.
Pi3K21B has been cloned and characterised.
