At the early third larval stage, there is a faint zfh2 signal in the centre of the leg discs, which is more readily visible at the middle of this stage, when it coincides with Dll expression. As the leg disc matures, several outer rings of zfh2 expression are successively observed. In accordance with the pupal ScerGAL4zfh2-LP30 expression in leg joints, strong zfh2 expression is observed in cells of everting pupal leg discs that will contribute to the formation of the joints, with more intense signal in cells abutting those forming the tarsal folds and weaker signal more proximally and distally in each tarsal segment. There is also strong signal in the pretarsus, the distal part of the leg, and in the leg tendon that reaches the pretarsus.
Markers that uniquely identify the cells of the NB3-7 lineage were used to examine the serotonin expressing cell lineages.
zfh2 is expressed in the lateral, but not the medial, of the paired ventro-lateral serotonergic neurons.
zfh2 protein isexpressed in about 50 cells/segment in the lateral embryonic CNS atpositions consistent with the subsequent location of serotonin cells. Inlarvae, it is expressed in a dispersed pattern in about 20% of the cellsin the ventral ganglion. A subset of these also express Ddc and includedopamine and seratonin neurons.
zfh2 protein is first detected in the CNS at the middle of germ band retraction. It is present in at least 50 neurons per hemisegment as well as in cells of the brain. It is also detected at lower levels in the hindgut of older embryos

Profile of zfh2 expression in the larval CNS shows intriguing overlap with Ddc expression in specific serotonin and dopamine neurons. The product of zfh2 binds to a protein binding site in the Ddc promoter.
The expression pattern of zfh2 suggests that the protein may be involved in neurogenesis.
